مرکبات منبع غنی متابولیتهای ثانویه هستند و نمو آن¬ها با تغییر در مقدار متابولیتهای اولیه و ثانویه همراه است. تلخی تأخیری مرکبات که در اثر عصارهگیری، یخزدگی، آسیبهای مکانیکی، انبارداری نامناسب و ... اتفاق می¬افتد یکی از مهمترین چالشهای فراوری مرکبات است. تلخی به ترکیبات لیمونوئیدی و فلاونوئیدی موجود در مرکبات برمیگردد که در دو مسیر بیوشیمیایی متفاوت سنتز میشوند. در مسیر لیمونوئیدی، لیمونات ای- رینگ لاکتون تولید شده و در ادامه دو مسیر مختلف را طی میکند. در مسیر اول آنزیم لیمونوئید دی-رینگ لاکتون هیدرولاز، لیمونات ای –رینگ لاکتون را به ماده تلخ لیمونین تبدیل میکند. در مسیر دوم لیمونات ای –رینگ لاکتون تحت تأثیر آنزیم لیمونوئید گلوکوزیل ترانسفراز گلوکوزیده شده و به ترکیبی غیر تلخ به نام لیمونین 17- بتا گلوکوپیرانوزید تبدیل میشود. در این مطالعه با نگاه همزمان به مسیر تلخی لیمونوئیدی مرکبات بیان ژن، پروتئومیک و متابولومیک تلخی لیمونوئیدی مرکبات در گونههای نارنگی (بهعنوان گونه شیرین)، پرتقال (بهعنوان گونه نسبتاً تلخ) و گریپ فروت (بهعنوان گونه تلخ) بررسی گردید. به همین منظور از بافت آلبدو و آبدانک هر سه گونه در زمانهای 20، 80، 110 و 200 روز بعد از تمام گل نمونهبرداری صورت گرفت و ارزیابی بیان ژن جهت بررسی رونوشتبرداری از ژن لیمونوئید گلوکوزیل ترانسفراز با استفاده از روشqRT- PCR انجام شد. همچنین مطالعه پروتئومیک با هدف بررسی الگوی پروتئوم تلخی در بافت با استفاده از الکتروفورز دو بعدی صورت پذیرفت و در نهایت پروتئینهای دارای تغییرات بیان تفسیر شدند. در ادامه نیز مطالعه متابولومیک جهت بررسی نوع و مقدار متابولیتهای موجود در بافت مایع با استفاده از HPLC انجام شد. این پژوهش نشان داد که بیشترین میزان بیان ژن (LGT) limonoid UDP-glucosyl transferas در پالپ گریپ فروت، نارنگی و پرتقال به ترتیب 200، 110 و 200 روز بعد از تمام گل بود. در مقایسه سه ژنوتیپ بیشترین میزان بیان ژن LGT در پالپ نارنگی ۱۱۰ روز بعد از تمام گل بود. بررسی بیان ژن LGT در آلبدو روندی شبیه پالپ داشت ولی میزان بیان این ژن در آلبدو کمتر از پالپ بود. Limonoate A ring lactone در گریپ فروت مشاهده نشد ولی در پرتقال و نارنگی ابتدا افزایش سپس کاهش نشان داد. Limonin 17 beta D glucopyranoside در گریپ فروت در ۲۰۰ روز بعد از تمام گل بهشدت افزایش نشان داد و در پرتقال و نارنگی روند صعودی از 20 روز بعد از تمام گل تا ۲۰۰ روز بعد از تمام گل نشان داد. در گریپ فروت در هر دو بافت پالپ و آلبدو تولید لیمونین روند افزایشی داشت. میزان تجمع لیمونین در پرتقال و نارنگی الگوی مشابه نشان داد به¬گونه¬ای که در اواسط مراحل رشدی افزایش و در انتهای رشد کاهش نشان داد اما الگوی در گریپ فروت الگویی کاملاً متفاوت با پرتقال و نارنگی مشاهده شد. آنالیز نتایج الکتروفورز دوبعدی شناسایی 121 لکه پروتئینی تکرارپذیر را درپی داشت. به ترتیب تعداد 19 و 17 لکه پروتئینی در گریپ فروت نسبت به پرتقال افزایش و کاهش بیان نشان دادند. به ترتیب تعداد 25و 25 لکه پروتئینی در گریپ فروت نسبت به نارنگی افزایش و کاهش بیان نشان دادند. همچنین به ترتیب تعداد 14 و 26 لکه پروتئینی در پرتقال نسبت به نارنگی افزایش و کاهش بیان نشان دادند. در بیان کلی نتایج پروتئومیک پژوهش حاضر، پروفایل پروتئینی متفاوت این سه ژنوتیپ را از یکدیگر نشان میدهد که با نتایج بررسی بیان ژن و بررسی متابولیت مطابقت دارد

Abstract: Citrus bitterness, which is divided into two categories: limonoid and flavonoid bitterness, is one of the major issues in the industry of this product. In the limonoid pathway, a substance called limonate A-ring lactone is produced and then goes through two different paths. In the first pathway, limonate A-ring lactone is converted to the bitter substance limonin by the enzyme limonoid d-ring lactone hydrolase, which causes a delayed bitterness in fruit extracts. In the second pathway, L-lactone is glucosidated by the limonoid enzyme glucosyltransferase to a non-bitter compound called 17-beta-glucopyranoside limonin. In this study, with a simultaneous look at the path of citrus limonoid bitterness with gene, proteomic and metabolic approaches, a comprehensive study of citrus limonoid bitterness in tangerine (as sweet) and orange (as relatively bitter) species and grapefruit (as a bitter species) was done. For this purpose, samples were taken from the albedo and pulp tissues of all three species at 20, 80, 110 and 200 days after full bloom. The expression of gene using the method qRT-PCR. Also, a proteomic study was performed by two-dimensional electrophoresis (in the first dimension by IEF method and in the second dimension by SDS-PAGE) and finally proteins with expression changes were interpreted. Subsequently, a metabolic study was performed to evaluate the type and amount of metabolites in fluid tissue using HPLC. In this study, with a simultaneous look at the path of citrus limonoid bitterness with gene, proteomic and metabolic approaches, a comprehensive study of citrus limonoid bitterness in tangerine (as sweet) and orange (as relatively bitter) species. And grapefruit (as a bitter species) was done. For this purpose, samples were taken from the albedo and Abdank tissues of all three species at 20, 80, 110 and 200 days after flowering and the expression of gene expression was evaluated to transcribe the limonoid gene of glucosyltransferase using the method. qRT-PCR was performed. Also, proteomic study was performed to investigate the pattern of bitterness proteome in tissue using two-dimensional electrophoresis (in the first dimension by IEF method and in the second dimension by SDS-PAGE method). This study showed that the highest expression of UDP-glucosyl transferas limonoid gene was in grapefruit pulp 200 days after flowering. The highest expression of UDP-glucosyl transferas limonoid gene was in mandarin pulp 110 days after flowering. The highest expression of UDP-glucosyl transferas limonoid gene was in orange pulp 200 days after flowering. Compared to the three genotypes, the highest expression of UDP-glucosyl transferas limonoid gene was in mandarin pulp 110 days after flowering. The expression of UDP-glucosyl transferas limonoid gene expression in albedo had a pulp-like process but the expression of this gene in albedo was less than pulp. In grapefruit, limonin production increased in both pulp and albedo tissues. The accumulation of limonin in oranges increased first in the middle of growth stages and decreased at the end of growth. Limonoate A lactone ring was not observed in grapefruit but in oranges and tangerines it first increased and then decreased. Limonin 17 beta D glucopyranoside in grapefruit showed a sharp increase in 3 days after flowering. Limonin 17 beta D glucopyranoside in oranges and tangerines showed an upward trend from 20 days after full flowering to 5 days after full flowering. The accumulation of limonin in tangerine increased first in the middle of growth stages and decreased at the end of growth. Accumulation of limonin in oranges and tangerines showed a similar pattern. However, the pattern of limonin accumulation in grapefruit was completely different from the pattern of oranges and tangerines. The proteomic results of the present study show the different protein profiles of these three genotypes from each other, which is consistent with the results of gene expression and metabolite studies.
