Thirteen factors thought to influence the release of protoplasts from leaf tissue of lemon (Citrus limon Bum.) were tested to develop a protocol to maximize protoplast yield. Maximal yields were achieved using leaves that had jut matured and were exposed to continuous light prior to and during the digestion period. A shaker speed of 100 rpm, pH of 5 and temperature of 29 1 C were the best physical parameters for leaf digestion. An osmoticum of 0.3M mannitol, 0.3M sucrose and 0.3M sorbitol was found to be better than any of the sugars individually. Different enzyme concentrations and combinations were tested and a mixture of 0.19% driselase and 0.51% pectinase was found to be most effective. Added salts and 2,4-dichlorophenoxy acetic acid had little effect on protoplast isolation.
