خشکی یکی از فاکتورصهای مهم محدود کننده تولید محصول در جهان و ایران است و این موضوع با افزایش تغییرات آب وص هوایی کره زمین اهمیت بیشتری پیدا میصکند .گندم غذای ضروری بیش از ۳۵ مردم دنیاست .بنابرین ارزیابی واکنش ارقام متفاوت گندم در برابر تنشصها ضروری است .اندازهصگیری آنزیم های آنتی اکسیدان در طول تیمارهای تنش خشکی به عنوان روشی جهت ارزیابی واکنش گیاه در برابر تنش پذیرفته شده است .در این پژوهش فعالیت آنزیم های آنتی اکسیدان و الگوی ایزوزیمی آنها در برگ و ریشهص گیاهچهصهای هشت رقم گندم پائیزه با حساسیت متفاوت به خشکی) چهار رقم حساس و چهار رقم مقاوم (در دو شرایط عادی و تنش مورد بررسی قرار گرفت .میزان بیان آنزیم ها با استفاده از نرمصافزار MCID اندازهصگیری شد .در گیاهچهصهای حاصل از بذرهای فوق دو ایزوفرم سوپراکسید دیسموتاز (SOD) و یک ایزوفرم کاتالاز (CAT) و هفت ایزوفرم پراکسیداز(POX) شناسایی شدند .یک ایزوفرم از SOD و دو ایزوفرم از POX بین ریشه و برگ گیاهچهصها متفاوت بودند .فعالیت CAT در تمامی ارقام در شرایط تنش بدون تغییر باقی ماند .اما SOD و برخی ایزوفرمصهای POX به طور معنیصداری در مقایسه با حالت عادی کاهش بیان پیدا کردند .ارقام متفاوت گندم پاسخصهای متفاوتی به تنش از خود نشان دادند .در برگ اختلاف معنیصداری (P<۰.۰۱)بین دو گروه حساس و مقاوم از لحاظ فعالیت کمی شده تمامی ایزوزیم های POX مشاهده شد، به طوری که ارقام مقاوم بیشترین فعالیت را به خود اختصاص دادند .اثر متقابل معنی داری بین تنش و ارقام گندم از لحاظ فعالیت کمی شده ایزوزیمصهایSOD ، CAT و POX در هیچکدام از بافت ها به دست نیامد .گروه مقاوم حدود ۳۸ افزایش فعالیت نسبت به گروه حساس برای POX کل نشان داد .در ریشهصها نیز همانند برگ ها SOD در شرایط تنش به شکل معنیصداری کاهش بیان نشان داد .ولی اثر متقابل شرایط محیطی و گروه-های گندم معنیصدار نبود .برای آنزیم CAT گروهصهای مقاوم گندم درحدود ۲۹ نسبت به گروه حساس افزایش بیان داشته و اختلاف معنیصدار نشان داد .چهارتا از ایزوزیم های پراکسیدازPOX۳ ،POX۴ ، POX۵ و POX۷ ریشه اثر متقابل، برای گروه گندم و تنش نشان دادند .به طوری که برای همه آنها در شرایط عادی میزان بیان آنزیم در گروه مقاوم و حساس از نظر آماری یکسان بود، ولی میزان بیان در شرایط تنش برای گروه مقاوم کاهش معنیصدار نشان داد .بنابراین از ایزوزیم های پراکسیداز ریشه در مطالعه پاسخ به تنشصها و نیز احتمالآ گزینش با کمک نشانگرص در گندم می توان استفاده کرد

Drought is one of the major factors limiting crop production globally and in Iran, with increasing global climate change making the situation more serious. Wheat is the staple food for more than 35 of world population, so assessment of different wheat cultivars's responses is essential. Measuring antioxidant enzymes during drought treatments is accepted as a way to evaluate plant's response to stress. In the present study, the responses of antioxidant enzyme activities and their patterns in root and leaf of seedlings of eight winter wheat cultivars differing in sensitivity to drought (four susceptible and four resistant cultivars) under normal and stress conditions were investigated. With MCID software expression rates of enzymes were evaluated. In seedlings above mentioned cultivars two isoforms of superoxide dismutase (SOD), one isoform of catalase (CAT) and seven isoforms of peroxidase (POX) were identified. One isoform of SOD and two isoforms of POX were different among leaves and roots. In all cultivars under stress, activity levels of CAT remained unchanged. Whereas expression of some peroxidase isoforms and SOD were significantly decreased as compared to control condition. Different wheat cultivars showed different responses to stress. In leaf, significant differences between susceptible and resistant groups were observed for densitometric activity of all peroxidases. So that resistant cultivars showed highest activity. Interaction effects of drought treatments and cultivars were not significant for activities of SOD, POX and CAT. Resistant group exhibited 38 increase in POX activities as compared to susceptible group. In root as well as leaves SOD expression showed significant decrease in stress condition but interaction of environmental conditions and genotype groups wasn't significant. Resistant group had 29 increase for CAT as compared to susceptible group and showed significant difference. Four isoforms of POX (POX3, POX4, POX5, POX7) showed highly significant interaction effect for wheat groups and stress in root. Nevertheless, for all of them in normal condition expression rates in both susceptible and resistant groups were similar statistically, but resistant group expression rate in stress condition showed significant decrease. So, use of root 's POX isozymes in study of responses to stress and also probably marker assisted selection of wheat would be usefull
