چکیده:اخیرا درمان زخم¬های عفونی به سبب ایجاد مقاومت آنتی بیوتیکی در باکتری¬ها به مشکل جهانی تبدیل شده است. سودوموناس آئروژینوزا به عنوان یکی از باکتری¬های دخیل در عفونت¬های پوستی مزمن دارای مقاومت چند دارویی بوده و تهدیدی برای سلامت عموم محسوب می¬شود. به همین سبب استفاده از روش¬های نوین ضروری به نظر می¬رسد. فاژ درمانی به عنوان یک روش کارآمد، دارای اثر ضد میکروبی قابل توجهی است. در این پژوهش هدف اصلی، ایجاد زخم پوش حاوی فاژ با قابلیت جلوگیری از عفونت باکتریایی و ترمیم سریع زخم بدون عارضه جانبی است.ابتدا سویه سودوموناس ائروژینوزا با تست¬های بیوشیمیایی مورد تایید قرار گرفت. سپس فاژهای متعددی علیه سودوموناس ائروژینوزا از آب¬های فاضلاب جداسازی و با استفاده از تایپینگ فاژی، دو فاژ پلی والانت برای تهیه کوکتل فاژی انتخاب شد. کوکتل فاژی جهت فاژ درمانی در هیدروژل تشکیل شده از پلیمرهای سدیم آلژینات و کربوکسی متیل سلولز بارگذاری شد. برای مقایسه اثر ضدمیکروبی، علاوه بر هیدروژل¬ حاوی فاژ، هیدروژل حاوی سیپروفلوکساسین، هیدروژل حاوی فاژ و سیپروفلوکساسین و هیدروژل خالص سنتز شد. اثر ضدمیکروبی هیدروژل¬ها به صورت invitro در محیط کشت و در شرایط invivo، در مدل موشی بررسی شد. هم¬چنین روند ترمیم زخم در گروه¬های مختلف موشی مورد ارزیابی قرار گرفت.نتیجه بررسی¬ها نشان داد که هیدروژل حاوی فاژ و هیدروژل حاوی آنتی بیوتیک اثر ضد میکروبی تقریبا همسانی دارند، اما از نظر روند ترمیم زخم و پاتولوژیک، عملکرد هیدروژل حاوی فاژ بهتر از آنتی بیوتیک است. استفاده از هیدروژل حاوی ترکیب فاژ و آنتی بیوتیک، از لحاظ اثر ضد میکروبی عملکرد بهتر و از نظر ترمیم زخم شبیه هیدروژل حاوی فاژ بود. با توجه به نتایج به دست آمده، هیدروژل حاوی فاژ با غیرفعال سازی سودوموناس ائروژینوزا و ترمیم سریع¬تر زخم، می¬تواند گزینه خوبی برای درمان زخم¬های عفونی باشد.

AbstractBackground: Recently, the treatment of infectious wounds has become a global problem due to the development of antibiotic resistance in bacteria. Pseudomonas aeruginosa, as one of the bacteria involved in chronic skin infections, has multidrug resistance and is a threat to public health. For this reason, the use of new methods seems necessary. Phage therapy as an effective method has a significant antimicrobial effect. The main purpose of this study was to create a phage-containing wound dressing with the ability to prevent bacterial infection and rapid wound healing without side effects.Methods: Pseudomonas aeruginosa strain was first confirmed by biochemical tests. Then, several phages against Pseudomonas aeruginosa were isolated from wastewater and two phage polyvalent phages were selected for phage cocktail using phage typing. Phage cocktails were loaded for phage therapy in a hydrogel composed of polymers of sodium alginate and carboxymethylcellulose. To compare the antimicrobial effect, in addition to phage-containing hydrogels, ciprofloxacin-containing hydrogels, phage-containing hydrogels and ciprofloxacin, and pure hydrogels were synthesized. The antimicrobial effect of hydrogels in vitro in culture medium and in invivo conditions, in addition to the antimicrobial effect, wound healing process in different groups of mice was also studied.Results: The results showed that phage-containing hydrogels and antibiotic-containing hydrogels have almost the same antimicrobial effect, but in terms of wound healing and pathological process, phage-containing hydrogels perform better than antibiotics. The use of a combination of phage and antibiotic hydrogels is better in terms of antimicrobial effect and similar to phage-containing hydrogels in terms of wound healing. According to the results, phage-containing hydrogels with inactivation of Pseudomonas aeruginosa and faster wound healing can be a good option for treating infectious wounds
