جو (Hordeum vulgare L. ssp. vulgare) یکی از مهمترین غلات پر مصرف با ارزش تغذیهصای قابل توجه در جهان به-شمار میصرود .جو پس از ذرت، برنج و گندم چهارمین محصول غلهصای با بیشترین میزان کشت در جهان و همچنین پنجمین محصول تولیدی مهم در جهان میصباشد .بیماریصهای برگی از مهمترین بیماریصهای جو در دنیا بهصشمار میصروند و باعث ایجاد خسارت اقتصادی روی جو در مناطق کشت و پرورش این محصول میصشوند .تحقیق حاضر در قدم اول با هدف شناسایی گونه-های قارچی مرتبط با بیماریصهای لکه برگی جو در استان آذربایجانصشرقی با استفاده از مفهوم تلفیقی گونه انجام گرفت .در مجموع ۵۴۰ جدایه قارچی از گیاهان جو و گرامینهصهای وحشی دارای علایم لکه برگی طی سال های۹۵ - ۹۶جداسازی و خالصصسازی گردید .براساس تلفیق دادهصهای ریختصشناختی با دادهصهای توالی ناحیهrDNA- ITSو gpdh تنوع قابل توجهی در گونهصهای قارچی مرتبط با بیماریصهای لکه برگی جو مشاهده گردید .در این تحقیق گونهصهایPyrenophora graminea ،P. teres ،P. semeniperda ، Bipolaris sorokiniana،B. salkadehensis ،Curvularia inaequalis ،C. spicifera ،C. neergaardii ،Exserohilum rostratum ،Rhynchosporium commune ، Preussia africana و Coniolariella hispanica شناسایی گردیدند .دو گونه P. graminea و P. teresبه ترتیب با فراوانی ۳۰/۶۱و ۸۵/۲۱ درصد، به عنوان گونهصهای قارچی غالب مرتبط با بیماریصهای لکه برگی جو در مناطق نمونه برداری شده، بودند .گزارش گونه P. semeniperdaبرای مایکوبیوتای ایران جدید است .جهت بررسی ساختار جمعیتی و تنوع ژنتیکی در جمعیتصهای گونه P. graminea در استان آذربایجانصشرقی از نشانگرهای مولکولی ISSR و RAPD استفاده شد .بررسی تنوع ژنتیکی با استفاده از سه آغازگر بین ریزماهوارهصای(DDB(CCA)۵ ، (AC)۸YA و (GA)۸C) و دو آغازگر تصادفی (AMER۶ و۱۷) - OPKصورت گرفت .با استفاده از پنج آغازگر) تلفیق دادهصهای ISSR و RAPD) از تعداد ۴۴ باند مورد تکثیر قرار گرفته تعداد ۴۰ باند (۹۱/۹۰ درصد (چندشکلی را نشانصداد .در این تحقیق میزان محتوای اطلاعات چندشکلی (PIC) بین ۱۳/۰ تا ۵۶/۰ متغیر و با میزان متوسط ۳۹/۰ بود .متوسط تنوع ژنی کل برابر با ۱۷۸/۰ بود که بین) ۰۸۷/۰ آذرشهر (و) ۲۷۵/۰ کلیبر (متغیر بود .میزان ضریب اطلاعات شانون بین ۱۲۶/۰ و ۵۶/۰ متغیر و با میزان متوسط ۲۷/۰ بود .تجزیهصی خوشهصای دادهصها در تلفیق دادهصهای نشانگرهایISSR و RAPD با استفاده از روش UPGMA و براساس ضریب تشابه دایس در سطح تشابه ۷۴ درصد جدایهصها را در ۲۰ گروه قرار داد .در تجزیه خوشهصای جدایهصها، گروهصبندی واضحی براساس منشا جغرافیایی جدایهصها مشاهده نگردید .فاصلهصی ژنتیکی بین جمعیتصها از ۰۱۱۱/۰ تا ۱۷۹۷/۰ متغیر بود .بیشترین فاصلهصی ژنتیکی بین جمعیتصهای میانه و کلیبر، کمترین فاصلهصی ژنتیکی بین جمعیتصهای مرند و شبستر دیده شد .کمترین و بیشترین میزان تمایز ژنی به ترتیب متعلق به جمعیتصهای ورزقان و خداآفرین (=(۰۳۵۹/۰Gst و جلفا و مرند با (=(۳۱۸۵/۰Gst بود .بنابراین بیشترین و کمترین میزان جریان ژنی به ترتیب در بین این جمعیتصها مشاهده گردید .براساس نتایج تجزیه واریانس مولکولیAMOVA ، بیشترین تنوع ژنتیکی در داخل جمعیت به میزان ۸۰ درصد و کمترین در بین جمعیتصها با میزان ۲۰ درصد با تمایز ژنتیکی قابل توجهی (۰۰۱/۰=P و (=۲۰۲/۰PhiPT ارزیابی گردید .بررسی اطلاعات حاصل از تنوع ژنتیکی جدایهصهای P. graminea مرتبط با لکه نواری بر جو و جو موشی، نشانصدهندهصی تطابق جدایهصها براساس میزبان ولی عدم تطابق با منطقه جغرافیایی بود .گونه P. graminea از مهمترین گونهصهای قارچی مرتبط با بیماری لکه نواری جو در جهان میصباشد .این گونه قارچی هتروتالیک میصباشد و در طبیعت براساس مرحلهصی غیرجنسی شناخته میصشود و اطلاعاتی از مرحلهصی جنسی فعال آن در درون جمعیتصهای این گونه در ایران و کشورهای تولید کنندهصی جو وجود ندارد .برای فهم امکان وجود مرحلهصی جنسی پنهان در درون جدایهصهای P. gramineaفراوانی و توزیع اللصهای تیپ آمیزشی در مقیاسصهای فضایی کوچک (microspatial) و بزرگ جغرافیایی (macrogeographical) بررسی و برآورد گردید .تعداد ۳۱۶ جدایهصی P. graminea جداسازی شده از گیاه جو از ۱۱۶ مزرعه واقع در ۵۳ منطقهصی جغرافیایی مختلف در استان آذربایجانصشرقی مورد بررسی قرار گرفت .در این تحقیق با بکارگیری روش واکنش زنجیرهصای پلیصمراز مولتیصپلکس توان تشخیص همزمان تیپ آمیزشی و شناسایی گونهصی P. graminea نیز فراهم گردید .در تمامی جدایهصهای مورد بررسیP. graminea ، باند اختصاصی - bp۴۳۵حضور داشت؛ همچنین یک قطعه - bp۱۳۰۰و یا یک قطعهصی - bp۱۱۵۰به ترتیب در جدایهصهای با تیپ آمیزشیMAT - ۱وMAT - ۲تکثیر گردید ۱۶۶ .جدایه متعلق به۱- MATو ۱۵۰ جدایهMAT - ۲بودند .نتایج این تحقیق نشان داد که فراوانی اللصهای تیپ آمیزشی در داخل و بین جمعیتصهای مختلف P. graminea توزیع تقریبا یکسانی را نشان میصدهد .= ;(۸۱۰/۰(۱:۱ ratio نتایج تعیین تیپصهای آمیزشی در مقیاس فضایی کوچک و در مقیاس بزرگ جغرافیایی نشانصدهندهصی حضور هردو تیپ آمیزشی اغلب در تمامی مقیاسصهای مورد مطالعه ازجمله درون یک زخم از یک برگ یک گیاه جو، مزارع یکسان، مزارع مختلف در یک منطقه و مناطق مختلف بود .براساس این تحقیق و بررسیصهایی که در گذشته بر روی ساختار جمعیت P. graminea صورت گرفته است میصتوان نتیجه گرفت که امکان وجود نوترکیبی جنسی در اکثر مناطق مورد تحقیق وجود دارد .در مطالعهصی حاضر روشصهای مختلف برای پی بردن به مرحلهصی جنسی شامل استفاده از قارچصکش، کاغذ صافی، لینولئیک اسید و کشت مخلوط برای تحریک مرحلهصی جنسی نتیجه بخش نبود

Barley (Hordeum vulgare L. ssp. vulgare) is one of the globally important cereal crops with significant nutritional value. After corn, rice and wheat, barely is ranked as the fourth highly cultivated cereal and also fifth most produced crop all over the world. Foliar diseases are considered among important barely diseases worldwide, causing economic crop losses in barely cultivation areas. Present study was aimed to identify fungal species associated with leaf spot disease of barley in East Azerbaijan province, using consolidated species concept. Totally 540 fungal strains were isolated and purified from barley and wild gramineous plants representing leaf spot symptoms during 2016-2017. Based on morphological and sequence data of ITS-rDNA and gpdh regions, a considerable variation was observed among the fungal species associated with barely leaf spot diseases. In this study, Pyrenophora graminea, P. teres, P. semeniperda, Bipolaris sorokinana, B. salkadehensis, Curvularia inaequalis, C. spicifera, C. neergaardii, Exserohilum rostratum, Rhynchosporium commune, Preussia africana, and Coniolariella hispanica were identified. P. graminea and P. teres were dominant species associated with leaf spot symptoms, with isolation frequencies of 61/30 and 21/85 percent, respectively. P. semeniperda is reported as a new taxon for the mycobioat of Iran. In order to understand the population structure and genetic variation of P.graminea population in East Azerbaijan Province, RAPD and ISSR molecular markers were applied using three microsatellite primers (DDB(CCA)5, (AC)8YA and (GA)8C) and two random primers (AMER6 and OPK-17). Using five primers (combined ISSR and RAPD data), 40 (90.91 ) polymorphic DNA bands were detected out of 44 different fragments amplified in this pathogen. The polymorphism information content (PIC) ranged from 0.13 to 0.56 with an average of 0.39. The average gene diversity was 0.178, and ranged from 0.087 (Azarshahr) to 0.275 (Kaleybar). The Shannons Information Index ranged from 0.126 to 0.406, with an average of 0.27. Cluster analyses using the unweighted pair-group method with arithmetic average (UPGMA) method and Dice coefficients for combined ISSR and RAPD markers data grouped P. graminea isolates into 20 clusters with similarity index 74 . No clear grouping according to geographical location of sampling was detected in cluster analysis. Genetic distances among all isolates ranged from 0.0111 to 0.1797. The greatest genetic distance was observed between Miyaneh and Kaleybar populations, while the lowest genetic distance was observed between Marand and Shabestar populations. The lowest and the highest level of genetic differentiation was observed between Varzegan and Khodaafarin (Gst = 0.0359) and Jolfa and Marand (Gst = 0.3185) groups, respectively suggesting the highest and the lowest level of cooperation (gene flow) between these populations, respectively. The analysis of molecular variation (AMOVA) indicated that most of the gene diversity (80 ) was distributed within populations, whereas 20 of the variation was found among populations and Significant genetic differentiation (PhiPT = 0.202, P = 0.001) was found among all populations. Based on genetic diversity assessment, there was a correlation between P. graminea isolates associated with leaf strip on barley and wild barley and their host while no correlation was observed between the isolates and their geographical origins. P. graminea is one of the main fungal species associated with barley leaf stripe disease worldwide. Even though a heterothallic mating strategy has been proven for P. graminea, this species is mainly known based on asexual morph in nature and there is not any available information on the prevalence of an active sexual cycle within the populations of this species in Iran as well as many other barley-producing countries. The feasibility of a cryptic sexual cycle within Iranian isolates of P. graminea was assessed through analyzing the distribution and frequency of the mating type alleles on microspatial and macrogeographical scales. A total number of 316 P. graminea isolates obtained from 116 fields in 53 geographical regions across East Azerbaijan province were assessed. A multiplex PCR assay was developed for simultaneous identification of P. graminea and screening of its mating type alleles using previously designed primer sets. In all studied P. graminea isolates, a 435-bp band was consistently amplified; while a 1300-bp fragment and a 1150-bp fragment was only amplified from the isolates harboring MAT-1 and MAT-2 alleles, respectively. The mating type identity of 166 isolates was determined as MAT-1 and 150 isolates as MAT-2. Results of the present study revealed nearly equal distribution (1:1 ratio; =0/810) of mating type alleles within and between different populations of P. graminea. Results of microspatial and macrogeographical distribution of mating types showed that both mating types were often present in almost all studied scales including, within the same lesion of each leaf from single barley plant and the same field, different fields in the same region and different regions. Based on the results of current study and referring to the earlier reports on the population structure of P. graminea, it is concluded that this pathogen undergoes regular cycles of sexual recombination in most of the examined regions. In the present study, the use of different methods to induce the sexual stage including fungicides application, use of filter paper, Linoleic acid and mixed culture was not efficient
