پیری برگ مرحلهصی نهایی فرآیندهای نموی یک برگ است که از نظر ژنتیکی برنامهصریزی صشده بوده و توسط برخی علامتصهای محیطی مانند شدت بالا یا کم نور و کمبود عناصر، القاء و تسریع میصشود .سلنیم عنصری ضروری برای گیاهان عالی محسوب نمیصشود با این وجود، برای رشد گیاهان بهصویژه در شرایط تنشی مفید است .گیاه کلزا (.(Brassica napus L با نیاز بالا به سولفور در مقایسه با سایر گیاهان زراعی، یک گیاه انباشتهصگر ثانوی سلنیم به شمار میصرود .در این پژوهش، تأثیر سلنیم بر پیری ناشی از قطع برگ، پیری نموی برگ و پیری القا شده با کمبود نیتروژن در گیاه کلزا مورد مطالعه قرار گرفت .گیاهان کلزا در شرایط اتاق رشد در بستر پرلیت آبیاری شده با محلول غذایی یا بهصصورت هیدروپونیک رشد یافتند و با سلنیم) بهصصورت سدیم سلنات (تیمار شدند .به منظور بررسی پیری القاء شده با قطع برگ، گیاهان ۷ هفتهصای به دو گروه تقسیم شدند :یک گروه قبل از قطع برگ با غلظتصهای۰ ،۱ ،۲ ،۵ ، ۱۰ و ۲۰ میکرومولار سلنیم پیشصتیمار شدند و در گروه دوم برگصها پس از قطع، با غلظتصهای۰ ،۲ ، ۱۰ و ۲۰ میکرومولار سلنیم تیمار گردیدند .برگصها در شدتصهای مختلف نور قرار گرفتند و SPAD برگ و شاخصصهای فلوئورسانس کلروفیل به مدت دو هفته تعیین شد .در پایان آزمایش، برگصهای تیمار نشده زرد و حتی نکروزه شدند در حالی که برگصهای تیمار شده با غلظتصهای بهینهصی سلنیم نسبتا سبز باقی ماندند .شاخصصهای مهم برگ از جمله فلوئورسانس کلروفیل نیز تأیید کنندهصی حفظ فعالیتصهای فتوسنتزی در برگصهای تیمار شده با سلنیم به مدت طولانیصتری، پس از قطع برگ بود .جهت مطالعهصی تأثیر سلنیم بر پیری نموی، برگصهای با سنین مختلف در گیاهان در سه مرحلهصی نموی مشخص) طوقهصای /ساقهصای-گلصدهی /میوهصدهی- پیری (بررسی شدند .تیمار سلنیم) به شکل سدیم سلنات (برای گیاهان شش هفتهصای با ۱۰ g سلنیم) به ازای گلدان (شروع شد، با ۵ g ادامه یافت و به مقدار کل ۷۵ g سلنیم) به ازای گلدان (در گیاهان برداشت شده در مرحلهصی میوهصدهی- پیری رسید .با افزایش سن برگ در طی سه دورهصی نموی، غلظت قند و پروتئین کاهش یافت، در حالی که با تیمار سلنیم افزایش پیدا کرد .همچنین فتوسنتز و تنفس تاریکی در حضور سلنیم افزایش یافت .تأثیر چشمگیر سلنیم بر شاخصصهای فلوئورسانس کلروفیل در برگصهای تحتانی مشاهده شد .سلنیم موجب تخصیص بیشتر نیتروژن به برگصهای پیر در مرحلهصی طوقهصای و ساقهصای-گل-دهی شد و بازانتقالی آن را کاهش داد، درحالیصکه در مرحلهصی میوهصدهی- پیری بازانتقالی آن به ساختارهای زایشی را تسریع کرد .در بررسی پیری القا شده با کمبود نیتروژن، تیمارهای مختلف نیتروژن شامل فراهمی کافی) شاهد، mM ۱۶) و کمبود نیتروژن (mM ۴ به مدت یک هفته و mM ۲ تا زمان برداشت (و تیمار سلنیم شامل بدون (Se) و با سلنیم (g Se ۷۵ به ازای گلدان (به صصورت ریشهصای و محلولصپاشی برگ برای گیاهان چهار هفتهصای اعمال شد .رشد گیاهان و تولید مادهصی خشک با کمبود نیتروژن کم شد و سلنیم موجب افزایش آنصها) بهصویژه در ریشه (گردید .فتوسنتز، شاخصصهای فلوئورسانس کلروفیل، غلظت رنگیزهصهای فتوسنتزی، قندها و پروتئینصهای محلول نیز با کمبود نیتروژن بهصویژه در برگصهای پیر کاهش یافت و در حضور سلنیم افزایش پیدا کرد .تیمار سلنیم هرچند باعث کاهش فعالیت سیستم دفاع آنتیصاکسیداتیو شد، موجب ایجاد تنش اکسیداتیو نیز نبود و حتی باعث کاهش آسیب ناشی از تنش نیز گردید .سلنیم بیان ژن رمز کننده پروتئین متصل به کلروفیل a/b (CAB) را افزایش و برعکس بیان ژن مهم پیری۲) - (SAG۱۲ ۱که رمز کننده نوعی سیستئین پروتئاز است را کاهش داد .با اینحال، کاهش در بیان این ژن با کاهش هماهنگ در فعالیت این آنزیم همراه نبود .نتایج این پژوهش نشان دهنده نقش سلنیم در مقابله با پیری برگ از طریق حفظ توانایی فتوسنتز، کاهش تأثیر عوامل تنش اکسیداتیو و نیز کاهش بیان ژنصهای پیری است

Leaf senescence is the terminal phase of developmental process of a leaf. Leaf senescence is a genetically programmed process, however, it can be induced and accelerated by external signals such as excess or low light conditions and nutrients deficiency. Selenium (Se) has not been considered as a necessary element for higher plants but it is beneficial for plant growth particularly under stress conditions. Oilseed rape (Brassica napus) with higher requirement for sulfur than other crops, is considered a secondary accumulator of Se. The objective of this Investigation was to investigate the ability of Se to counteract leaf senescence in oilseed rape. In addition of developmental senescence, nitrogen (N) deficiency and leaf detachment were utilized to study the progress of leaf senescence as well as Se effect on this process. Oilseed rape plants were grown under growth chamber conditions in perlite irrigated with nutrient solution or hydroponically and treated with Se as sodium selenite (Na2SeO4). For study of Se effect on the senescence of detached leaves, 7 weeks after sowing plants were divided into two groups; the first group was pretreated before leaf detachment with 6 levels of Se (0, 1, 2, 5, 10, and 20 M) for one week and leaves of the second group were treated after leaf detachment with four levels of Se (0, 2, 10, and 20 M). Leaves were placed under different light intensities and leaf SPAD and fluorescence parameters were determined for two weeks. At the end of experiment, detached leaves turned yellow or even became necrotic while Se-treated leaves remained fairly green 3 weeks after detachment. Leaf chlorophyll fluorescence parameters confirmed maintenance of photosynthetic capacity of detached leaves for longer time in both Se-treated and pre-treated leaves. In order to investigate the selenium effect on developmental senescence, leaves of different age belonging to the plants at three distinct developmental stages (rosette, bolting-flowering and fruiting-senescence) were harvested and analyzed. Se treatment was started for six-week-old plants with 10 g pot1, followed by 5 g Se pot1 and reached the total amount of 75 g Se pot1 in the plants harvested at fruiting-senescence stage. The total carbohydrate and protein concentrations of leaves decreased with leaf age, while increased by the Se treatment. Photosynthesis and dark respiration rates were both elevated by Se. Significant effect of Se on chlorophyll fluorescence parameters were observed on the lower leaves. At rosette and bolting stages, Se increased nitrogen allocation to the old leaves and diminished its remobilisation, whereas accelerated nitrogen remobilisation towards reproductive parts at fruiting stage. For study of N deficiency-induced leaf senescence, different N treatments including adequate (control, 16 mM) and low N were supplied for four-week old plants. Low N treatment was started with 8 mM (for two weeks) followed by 4 mM (for one week), and finally was applied at 2 mM throughout the further growth period until harvest. Se treatments were started as root application or leaf spray simultaneous with N treatments. Plants growth and dry matter production decreased with N deficiency, and Se increased it particularly in the roots. Furthermore, photosynthesis, chlorophyll fluorescence parameters, concentrations of photosynthetic pigments, sugars and proteins decreased with N deficiency, particularly in the old leaves while increased in the presence of Se. Although Se reduced the activity of antioxidative enzymes, it did not cause oxidative stress and rather reduced damage of stress. Gene expression analysis of the chlorophyll a/b binding protein (CAB) and SAG12 1-2 (Senescence-Associated-Genes 12 1-2) encoding a cysteine protease, revealed that Se increased the expression of CAB while decreased the expression of SAG12 1-2 in the senescing leaves. The latter, unexpectedly, was not accompanied by lower activity of cysteine protease. Our results indicated that, Se delays leaf senescence through maintenance of photosynthetic activity, reduction of oxidative stress and down-regulation of senescence-associated genes
