امروزه روغن سیاهدانه به علت داشتن ترکیبات زیست فعال متعدد مورد توجه ویژه قرار گرفته است .از آنجا که روشهای استخراج بر روی ترکیبات زیست فعال اثر گذار هستند، در این پژوهش، اثر پرس سرد و پرس گرم بر روی ترکیبات زیست فعال و کیفیت روغن سیاهدانه مورد بررسی قرار گرفت .روغن با دو روش پرس سرد (T۱) در دمای محیط و پرس گرم (T۲) در دمای ۱۰۰ درجه سانتیگراد از دانه سیاهدانه استخراج و سپس آزمایشهای کیفی طی روزهای۱ ،۳۰ ، ۶۰ و ۹۰ انجام شد .درصد استخراج روغن تیمار( T۲ (۲۳ ، بیشتر از تیمار ( T۱ (۲۱ بود .عموما، عدد اسیدی روغنهای استخراجی از هر دو روش مشابه بودند و در طی نگهداری افزایش معنیدار (۰۵/۰> p) یافتند .اندیس پروکسید تیمار T۱ طی مدت زمان نگهداری روند افزایشی را نشان داد اما تیمار T۲ تا روز۳۰ ، روند افزایشی و سپس روند کاهشی را در پی داشت .مقادیر کلروفیل، تیمار T۱ و T۲ تا روز۶۰ ، کاهش معنیداری داشتند و سپس در هر دو تیمار، کلروفیل در روز ۹۰ افزایش یافت، اما به طور کلی مقدار رنگدانه کلروفیل تیمار T۲ نسبت به تیمار T۱ بیشتر بود .مقدار کاروتنوئید در تیمارT۱ و T۲ طی ۹۰ روز نگهداری کاهش یافت .ترکیبات فنلی تا روز۶۰ ، در هر دو تیمار کاهش معنیداری یافت اما در روز۹۰ ، ترکیبات فنلی هر دو تیمار افزایش یافت، هرچند در طی نگهداری تیمار T۲ نسبت به تیمار T۱ ترکیبات فنلی بیشتری داشت .نتایج رنسیمت نیز نشان داد که روغن حاصل از پرس گرم پایداری اکسیداسیون بیشتری نسبت به روغن حاصل از پرس سرد داشت .در کل با توجه به نتایج بدست آمده میصتوان بیان کرد که روغن سیاهدانه استخراجی با روش پرس سرد و پرس گرم به علت داشتن اسیدهای چرب غیراشباع بالا و ترکیبات زیست فعال متعدد به عنوان یک روغن خوراکی با خواص تغذیهای، دارویی و درمانی بسیار مفید میصباشد، ولی با توجه به مقدار کلروفیل بالای آن و افزایش سریع عدد پروکسید روغن حاصله از هر دو روش، باید در استخراج روغن و در طی نگهداری روغن تدابیر ویژهصای) نگهداری در مکان تاریک و خنک (صورت گیرد تا کیفیت روغن حفظ شود و ترکیبات مضر همچون پروکسیدها و اسیدهای چرب آزاد کمتر تولید شوند

Nowadays black seed (Nigella sativa) oil has been highlighted for its special bioactive compounds. As extraction procedures are effective on bioactive compounds of black seed oil, cold and hot press effect on bioactive compounds of black seed oil has been studied in this study. The oil has been extracted from black seed with cold press (T1) on room temperature and hot press (T2) on 100 C and then the extracted oil has been quality tested during the days of 1, 30, 60 and 90. The percentage of extraction of T2 treatment oil (23 ) was higher than that of T1 treatment (21 ). Generally, the acid number of extracted oils was similar in both methods and during storage, a significant increase (p < 0/05) Found. The T1 peroxide value showed an increasing trend during storage, but the T2 treatment had an increasing value until day 30 and then a decreasing value. Chlorophyll of T1 and T2 treatments decreased significantly till day 60 and then in both treatments, chlorophyll was increased on day 90, but in general, the amount of T2 chlorophyll chromo was higher than T1 treatment. The amount of carotenoids in the T1 and T2 treatments decreased during the 90 days storage. Phenolic compounds significantly decreased in both treatments until day 60, but on day 90, phenolic compounds of both treatments increased, although during storage T2 treatment, more phenolic compounds were present than T1 treatment. Rancimat results also showed that the oil from the hot pressing had more oxidation stability than the cold pressing. In general, according to the results, it can be concluded that black seed oil extraction from cold press and hot press is very useful due to unsaturated fatty acids and numerous bioactive compounds as an edible oil with nutritional, medicinal and therapeutic properties However, due to its high chlorophyll content and a rapid increase in the amount of oil peroxide, there is a need to special attention during extraction and storage of oil (storage in dark and cold place) to maintain the quality of the oil and harmful compounds such as proxides and free fatty acids are produced less
