کیتیناز از جلد Ephestia kuehniella Zeller (Lepidoptera: Pyralidae) استخراج و سپس با استفاده از روش کروماتوگرافی تعویض کاتیونی روی ستونsepharose - SPخالصصسازی شده و ویژگیصهای آن بررسی گردید .وزن مولکولی پروتئین توسط الگوی الکتروفورز ژل پلیصاکریلصآمید احیایی یا واسرشتصگرPAGE) - (SDSمقدار ۷/۷۹ کیلو دالتون تعیین شد .وزن مولکولی کیتیناز E. kuehniella به کیتیناز سایر حشرات شباهت داشت .بهینه اسیدیته ۶ و دمای فعالیت بهینه آنزیم ۴۰ درجه سلسیوس تعیین گردید .آنزیم در اسیدیته ۵/۴ تا ۸ بهصطور وضوح ثبات نشان داد .در میان یونصهای فلزی سنجیدهصشده، با توجه به تاثیر آنها روی فعالیت آنزیمی، آنزیم خالص شده بیشترین حساسیت را به یونصهای کبالت، منیزیوم، کلسیم، جیوه و نقره نشان داد .مقادیر Km و Vmaxبرای کیتیناز E. kuehniella نسبت به کیتین کلوئیدی به عنوان زیرنهشت به ترتیب ۹۸/۳ میلی گرم بر میلی لیتر و ۱۱/۷ میکرومول بر دقیقه بر میلی گرم بودند .در راستای شناسایی ژن کتیناز بیدآرد DNA ژنومی حشره بالغ تهیه شد .برای تکثیر DNA کیتیناز، DNA جدا شده از حشره توسط یک جفت پرایمر دژنرهصی طراحی شده قبلی در واکنش زنجیرهصای پلی مراز به کار رفت .محصول حاصل حدود ۵۳۵ جفت باز مطابق با قطعه قابل پیش بینی بود .این محصول با ناحیهصی همتای رمز کننده کیتیناز در سایر حشرات مقایسه شد .در نتیجه، بیشترین شباهت به کیتیناز پروانهصها تشخیص دادهصشد .ویژگیصهای کیتین بیدآرد در مقایسه با کیتین تجاری از میگو مقایسه گردید .چنانکه فعالیت ویژه کیتیناز بیدآرد مستقیما با ویژگیصهای فیزیکی-شیمیایی کیتین به عنوان زیرنهشت مرتبط است .نتایج نشان داد که بدن بیدآرد مقادیر قابل توجهی کیتین و در مقابل دارای مقادیر کمی مواد معدنی است .بررسیصهای طیف سنجی مادون قرمز تبدیل فوریهIR)- (FT، تجزیه عنصری، تصویرصبرداری میکروسکوپ الکترونی و تفرق انرژی انکسار اشعه ایکسEDX) - (SEMجهت بررسی مشخصات فیزیکی کیتین استفاده شدند .طیفIR - FTنشان داد که کیتین بیدآرد با توجه به جهت میکروصفیبرهای خود دارای ساختار آلفاصی ()کیتین است .درجه استیله شدن کیتین بیدآرد و کیتین تجاری با استفاده از روشIR - FTبه ترتیب ۴۲/۷۱ و ۸۲/۷۰ درصد محاسبه شد .تصاویر میکروسکوپ الکترونی وجود ساختار مواد فیبری و منافذ ریز را در سطح نمونهصهای کیتین نشان داد .اندازه منافذ وعرض نانوفیبرهای کیتین بیدآرد به ترتیب حدود۲/۵ ۳/۱میکرومتر و۹۵ ۸نانومتر بود .مطالعه فعالیت آنزیمی نسبت به کیتین کلوئیدی به عنوان زیرنهشت نشان داد که کیتین بیدآرد خلوص شیمیایی بالایی برابر خلوص کیتین تجاری دارد .تجزیه کوتیکول حشرات ماده E. kuehniella توسط کروموتوگرافی گازی و طیف سنجی جرمی وجود چهار گروه ترکیبات شیمیایی شامل آلکان، الکل، آلدئید واستر را نشان داد .آلکانصهای شناسایی شده، شامل زنجیرهای کربن با تعداد ۱۲ تا ۱۸ ،۲۰،۲۳ ، ۲۴ و ۲۹ اتم کربن بودند .بیشترین فراوانی را زنجیر ۱۴کربنی با ۹۸/۱۴ درصد و ۱۸ کربنی با ۱۵/۸ درصد داشتند .اسیدصهای چرب شامل هگزاصدسنوئیک اسید(:(C ، اسید اولئیک (:(C و اسید لینولئیک (:(C بودند .الکلصهایoctadecadienol (C:) -۳,۱۳-Z,Z-Methyl- ۲وol acetate (C:) -۱-tetradecen-۱۰-Z-Methyl- ۹در لیپید کوتیکولی بیدهای جنس ماده مشاهده شد .دو آلدئید۱۶:Ald) -hexadecenal (E۱۱-۱۱- (E)و۱۸:Ald) -Octadecadienal (Z۹,۱۷-۹,۱۷- (Z)در عصاره استخراج شده بیدآرد شناسایی گردید .اثرات آنتی بیوتیکی کیتیناز و ترکیبات بیوشیمیایی کوتیکولی استخراج شده از جلد بیدآرد در مقابل باکتریصهای گرم مثبت و گرم منفی سنجش شد .کیتیناز رشد تمام باکتریصهای آزمایش شده را مهار کرد .در حالی که ترکیبات کوتیکول بیدآرد فقط رشد باکتریصهای گرم مثبت را مهار کردند

Chitinase from Ephestia huehniella Zeller ( Lepidoptera: Pyralidae) integument was extracted and then purified by cation exchange chromatography on SP-sepharose before characterization. Its molecular mass was determined to be 79.7 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). E.kuehniella chitinase had similar properties to other insect chitinases with respect to its molecular mass. The optimum pH and temperature for its activity were 6 and 40C, respectively. The enzyme exhibited obvious pH stability from 4.5 to 8. Among the metals that were assayed in regard to their influence on the enzyme activity, the purified enzyme showed high sensitivity to Co, Mn, Ca and Ag. The Km and Vmax values of the chitinase were 3.98 mg mL and 7.11 mol min mg for colloidal chitin, respectively. With respect to identification of chitinase gene from the moth, total DNA was prepared from the adults. The isolated DNA was subjected to PCR using a pair of the previously designed-degenerate primers. The obtained product was about 535 base pairs corresponding to the anticipated fragment. The product was compared to counterpart chitinase coding region of other insects. As a result, the highest similarity was identified to the lepidopteran chitinase. The moth chitin characteristics were compared with shrimp chitin because the specific activity of the moth chitinase is directly associated with physicochemical properties of chitin as substrate. The results revealed that the moth body contained significant amounts of chitin but lower amounts of minerals. Fourier transform infrared spectroscopy (FT-IR), elemental analysis (EA), scanning electron microscopy (SEM) and energy dispersive x-ray diffraction (EDX) were used to investigate physical characteristics of the chitin samples. FT-IR spectra revealed that the moth chitin had -chitin form (anti-parallel chains) with respect to the different orientations of its microfibrils. The degree of acetylation of chitin from moth and shrimp calculated using the FT-IR stretching bands were 70.82 and 71.42 , respectively. The SEM observation exhibited the presence of fibrillar material and porous structures in surface of the chitin samples. The size of the pores and the width of the nanofibers of the moth chitin were in the range of 5.21.3 m and 958 nm, respectively. The study on enzymatic activity toward colloidal chitin as substrate showed that the extracted chitin from the moth had high chemical purity as good as that of commercial chitin. Cuticular analysis of E. kuehniella females by gas chromatography-mass spectrometry revealed the presence of four groups of chemical compounds which are alkane, alcohol, aldehyde and ester. Cuticular n- alkanes ranged from 12-18, 20, 23, 24 and 29 carbon atoms in the chain. Cuticular fatty acids included hexadecenoic acid, 9-octadecenoic acid and 9,12- octadecenoic acid. Two alcohols, 2-Methyl-Z,Z-3,13-octadecadienol and 9-Methyl-Z-10-tetradecen-1-ol acetate, were found in the cuticular lipids of females. Two aldehyde components, (E)-11-hexadecenal and 9,17-octadecadienal, were identified in the cuticular extract of the moth. Antibacterial effect of the moth chitinase and cuticular compounds on both Gram positive and Gram negative bacteria were assayed. As a result, the enzyme inihibited growth of all of the tested bacteria. However, the compounds from the moth cuticle inhibited growth of only Gram-positive bacteria
