Breast cancer is the leading cause of death in women globally and the current treatments have several adverse effects and may lead to resistance. Plant-derived metabolites may offer efficient and less harmful anticancer agents. Hesperetin (HSP) is an herbal flavonoid with anti-tumor capabilities, and it has been investigated as a potential treatment for different types of cancer. This study aimed to investigate the cytotoxic and anti-malignant potential of hesperetin on breast cancer (BT-474) and normal cells (MCF-10a) by analyzing the effect of seven different concentrations of hesperetin using MTT assay, real-time PCR and, flow cytometry. We investigated the cytotoxicity, apoptotic effect, and MLH1/MSH2 gene expression of HSP-treated breast cancer cell lines including the cancerous cell line BT-474 and normal cell line MCF-10-A. Our results showed that HSP has dose-dependent cytotoxicity in BT-474 rather than MCF-10A cells. The higher the concentration of HSP, the lower the cell viability and proliferation. The IC50 of HSP in BT-474 cancer cells after a 48-hour exposure was 279.2 µM, while the IC50 in normal cell lines during the same exposure period was 855.4 µM. HSP was able to inhibit the proliferation of the BT-474 cell line at different IC50 concentrations within 48 hours. The cytotoxicity of HSP was more significant in cancer cell lines than in normal cells, which is a positive factor for using the drug to treat breast cancer patients. The apoptotic effect of HSP in BT-474 cells was also investigated, and it was found that the higher the concentration of HSP, the more the cells underwent apoptosis. Furthermore, we found that the highest concentration of HSP led to overexpression of the MLH1 and MSH2 genes in both breast cancer and normal cell lines. Overall, our study suggests that HSP has an anticancer effect on breast cancer cell lines, and the effect is concentration-dependent.

سرطان پستان علت اصلی مرگ و میر زنان در سراسر جهان است و درمانهای موجود دارای اثرات جانبی منفی هستند و ممکن است مقاومت ایجاد کنند. متابولیتهای بهوجود آمده از گیاهان میتوانند درمانهای ضدسرطانی موثر و کمسمیتتری را فراهم کنند. هسپرتین (HSP) یک فلاونوئید طبیعی با خواص ضدتوموری است که به عنوان یک ترکیب درمانی برای سرطان مورد تحقیق قرار گرفته است. با مطالعه اثرات هفت غلظت مختلف هسپرتین با استفاده از آزمون MTT، PCR Real-time و فلوسیتومتری، سمیت سلولی، اثر آپوپتوزی و بیان ژنهای MLH1 و MSH2 را بررسی نمودیم و در طول این مطالعه تاثیر هسپرتین بر لاین های سلولی سرطانی پستان BT-474 و طبیعی MCF-10a را بررسی کردیم. یافتههای ما نشان داد که HSP در یک روش وابسته به غلظت، در سلولهای BT-474 نسبت به MCF-10A سمیت سلولی بیشتری ایجاد میکند. بقای سلولها و تکثیر آنها با افزایش مقادیر HSP کاهش مییابد. پس از تیمار به مدت 48 ساعت، IC50 HSP در سلولهای سرطانی BT-474 برابر با 279.2 M/ml بود، در حالی که IC50 برای سلولهای طبیعی 855.4 M/ml بود. HSP در مدت 48 ساعت توانست در دوزهای مختلف از تکثیر لاین سلولی BT-474 جلوگیری کند. بررسی تأثیر آپوپتوتیک HSP در سلولهای BT-474 نشان داد که میزان آپوپتوز با غلظت HSP افزایش مییابد. همچنین، یافته ها نشان دادند که در هر دو لاین سلولی سرطان پستان و طبیعی، ژن های MLH1 و MSH2 در هنگام حضور HSP به میزان بیشتری بیان میشوند. به طور کلی، پژوهشهای ما نشان میدهد که HSP تأثیر ضدسرطانی وابسته به غلظتی روی لاین سلولی سرطان پستان دارد.
