5 در 86 بیمار تالاسمی که ژنوتیپ بتا گلوبین هموزیگوت و یا `HS4 Beta Globin LCR ناحیه ی هتروزیگوت مرکب داشتند بررسی شد .با مقایسه ی داده های بدست آمده از این مطالعه و داده های موجود از 5`Beta Globin در G سایر نواحی مرتبط در جمعیت بیمار و 101 فرد نرمال مشخص شد که فرکانس آلل مثبت در بیماران تالاسمی نسبت به جمعیت نرمال بالاتر است .ناهمسنگیHBG - Xmn1 و LCR HS4 2 مشاهده شد .فرکانس فنوتیپ کلینیکیIVSII - مثبت و جهشHBG2 - 1 Xmn1 ، Gپیوستگی بین آلل داشتند نسبت به/++ GG 5 ژنوتیپHBG - `HS4/Xmn1خفیف تر در بیمارانی که در لوکوس های 2 داشتند بیشتر بود .تمامی این مطالب به همراه شواهد /AA بیمارانی که در همین لوکوس ها ژنوتیپ در بیان ژن گلوبین هستند که درHBG 5 - Xmn1و `HS 2 بیوشیمیایی، حاکی از نقش پلی مورفیسم های 4 بر بیان ژن گاما گلوبین چنین مطرح می کنندHBG - Xmn1غیاب مکانیسم ها یا مسیر های نحوه ی تاثیر 2 در رابطه با افزایش بیان ژن گاماگلوبین نسبت داده می شدHBG - Xmn1که شاید نقشی که مدتها بود به 5 2 ایفا می شود `HS .توسط عوامل پیوسته با آن از جمله پلی مورفیسم 4 کلمات کلیدیIVSII - 1،HBG2 -Xmn1،Globin LCR 5`HS -Beta بتا تالاسمی،4
HS4 palindromic site on modifying chromatin structure and in the absence of any evidence from functional studies relating the Xmn1-HBG2 site to the increased gamma chain expression, suggest that the phenotype modifying role long time assigned to Xmn1-HBG2 is possibly played by more functionally potent elements linked to it in LCR؟۱HS4/Xmn1-HBG2 loci. These data together with biochemical evidence suggesting a role for the A/G polymorphism at 5؟۱HS4. The frequency of milder clinical phenotype was significantly higher in patients with GG/++ than cases with AA/. . genotypic pattern in 5؟۱HS4 (TGGGG A/G CCCCA) and positive Xmn1-HBG2 profile was significantly higher in thalassemia patients compared to the normal population. Linkage disequilibrium was observed between the G allele and positive Xmn1-HBG2 profile in patient population. Furthermore, dominance of IVSII-1 in the mutation spectrum of the patients enabled us to identify linkage disequilibrium relationships between IVSII-1, positive Xmn1-HBG2 and the G allele at 5؟۱The core sequence of 5`HS4-beta globin locus control region was analyzed in 86 thalassemia patients with homozygous or compound heterozygous beta globin gene mutations, along with Xmn1-HBG2 genotype and other related regions compared with equivalent regions in 101 normal individuals.
