درخت هلو (Prunus persica) یکی از مهم¬ترین درختان میوه هسته¬دار است که دارای گلدهی زود هنگام بوده و با توجه به وجود سرماهای دیر رس بهاره در مناطق معتدله و حساسیت گل¬های هلو نسبت به این عامل گاهی خسارت¬های قابل ملاحظه¬ای مشاهده می¬شود. در این راستا آزمایشی به منظور ارزیابی امکان کاهش خسارت ناشی از سرمای دیررس بهاره با استفاده از تیمار ملاتونین بر روی دو رقم درخت هلو انجام شد. آزمایش به صورت فاکتوریل در قالب طرح کاملا تصادفی با سه غلظت از ماده ملاتونین (صفر، 100و 200 میکرو¬مولار) و سه تیمار دمایی (5/1-، 5/2- و 5/3- درجه سانتی¬گراد) با سه تکرار انجام شد. 12 ساعت بعد از استعمال ملاتونین، شاخه¬های تیمار شده تحت تیمار سرمایی قرار گرفت. پس از سه ساعت قرار گرفتن در تیمار سرمایی، شاخه¬ها به دمای اتاق منتقل و بعد از 24 ساعت خسارت مادگی بررسی شد. سپس ویژگی¬هایی مانند میزان فعالیت آنزیم¬های آنتی اکسیدانی (سوپر اکسید دیسموتاز، کاتالاز و پراکسیداز)، میزان پرولین و پرتئین کل مورد ارزیابی قرار گرفت. نتایج بررسی مادگی نشان داد کاربرد ملاتونین در غلظت 200 میکرو¬مولار در دمای 5/1- درجه سانتی¬گراد در هر دو رقم، باعث شد تخمدان کمتر خسارت ببیند. بیشترین درصد کلاله¬های خسارت دیده مربوط به تیمار شاهد در رقم مخملی در دماهای 5/2- و 5/3- درجه سانتی¬گراد بود. کمترین درصد کلاله¬های خسارت دیده در تیمار¬های که از غلظت 200 میکرو¬مولار ملاتونین استفاده شده بود دیده شد. بیشترین مقدار پرولین مربوط به تیمار ملاتونین 100 میکرو¬مولار در دمای 5/2- درجه سانتی¬گراد و کمترین مقدار آن مربوط به تیمار 200 میکرو¬مولار ملاتونین در دمای 5/3- درجه سانتی¬گراد در رقم حاج کاظمی بود. بیشترین مقدار پروتئین مربوط به تیمار200 میکرو¬مولار ملاتونین در دمای 5/1- درجه سانتی¬گراد بود و کمترین میزان پروتئین مربوط به تیمار 200 میکرو¬مولار ملاتونین در دمای 5/3- درجه سانتی¬گراد مشاهده شد. در بررسی فعالیت آنزیم SOD بیشترین میزان فعالیت این آنزیم در رقم مخملی در تیمار 100 میکرو¬مولار ملاتونین در دمای 5/3- درجه سانتی¬گراد به دست آمد. کمترین میزان فعالیت آنزیم SOD مربوط به تیمار 100 و 200 میکرو¬مولار در دمای 5/2- درجه سانتی¬گراد در رقم حاج کاظمی بود. بیشترین میزان فعالیت آنزیم CAT مربوط به رقم حاج کاظمی با 100 میکرو¬مولار ملاتونین در دمای 5/2- درجه سانتی¬گراد و کمترین مقدار آن مربوط به تیمار 200 میکرو¬مولار در دمای 5/2- درجه سانتی¬گراد بود. بیشترین فعالیت آنزیم POD در رقم مخملی به دست آمد که مربوط به تیمار ملاتونین 100 میکرو¬مولار تحت تاثیر دمای 5/3- درجه سانتی¬گراد بود و کمترین مقدار آن در تیمار 200 میکرو¬مولار ملاتونین در دمای 5/2- درجه سانتی¬گراد در رقم حاج کاظمی مشاهده شد.
AbstractThe peach tree (Prunus persica) is one of the most important stone fruit trees that has early flowering and due to the presence of late spring colds in temperate regions and the sensitivity of peach blossoms to this factor, considerable damages are sometimes observed. In this regard, an experiment was conducted in order to evaluate the possibility of reducing damage caused by late spring cold by using melatonin treatment on two varieties of peach trees. The experiment was conducted as a factorial in a completely randomized design with three concentrations of melatonin substance (zero, 100 and 200 μM) and three temperature treatments (-1.5, -2.5 and -3.5 degrees Celsius) with three repetitions. 12 hours after using melatonin, the treated branches were subjected to cold treatment. Then, characteristics such as antioxidant enzyme activity (superoxide desmutase, catalase and peroxidase), the amount of proline and total protein were evaluated. The results of the examination of the female showed that the use of melatonin at a concentration of 200 µM at a temperature of -1.5 °C in both cultivars caused less damage to the ovary. The highest percentage of damaged stigmas was related to the control treatment in the Makhmali variety at temperatures of -2.5 and -3.5 degrees Celsius. The lowest percentage of damaged stigmas was seen in the treatments that used a concentration of 200 µM melatonin. The highest amount of proline was related to the treatment of 200 μM melatonin at a treatment of -1.5°C and the lowest amount of protein related to the treatment of 200 μM melatonin was observed at the treatment of -3.5°C in Haj Kazemi variety. The highest amount of protein related to the treatment of 200 μM melatonin was observed at the temperature of -1.5°C and the lowest amount of protein related to the treatment of 200 μM melatonin was observed at the temperature of -3.5°C. In the study of SOD enzyme activity, the highest level of activity of this enzyme was obtained in the velvet variety in the treatment of 100 μM melatonin at -3.5°C. The lowest level of SOD enzyme activity was related to the treatment of 100 and 200 µM at -2.5°C in Haj Kazemi variety.In examining the activity of SOD enzyme, the highest activity of this enzyme was obtained in the Makhmaly variety in the treatment of 100µM melatonin at -3/5°C. The lowest level of SOD enzyme activity was related to the treatment of 100and 200 µM at -2/5°C in Haj Kazemi cultivar. The highest amount of CAT enzyme activity was related to Haj Kazemi cultivar with 100 micromolal melatonin at -2.5°C and the lowest value was related to 200 micromolar treatment at -2.5°C. The highest activity of POD enzyme was obtained in the Makhmali variety, which was related to the treatment of 100 μM melatonin under the influence of temperature -3.5 °C, and the lowest value was observed in the treatment of 200 μM melatonin at the temperature of -2.5 °C in Haj Kazemi cultivar.
