بیماری پوسیدگی نرم (Soft-rot) در ایران و بیشتر نقاط دنیا اهمیت اقتصادی فراوانی دارد. سیب زمینی و هویچ از حساسترین میزبانهای این بیماری هستند و هر ساله خسارت قابل ملاحظهای ناشی از این بیماری روی این دو محصول گزارش میشود. بسیاری از گونه¬های جنس Pectobacterium عامل پوسیدگی نرم گزارش شده¬اند. دو زیر گونه Pectobacterium carotovorum subsp. carotovorum (Pcc) و(Pco) Pectobacterium carotovorum subsp. odoriferum از مهم-ترین و شایع¬ترین عوامل مولد بیماری هستند. از گذشته¬های دور، خاصیت باکتری¬کشی مس جهت کنترل بیمارگر¬های مختلف به کار رفته است اما با توجه به آثار نامطلوب زیست محیطی ناشی از تجمع مس در گیاهان، خاک، آب و زنجیره¬های غذایی تلاش بر این است که مصرف آن به حداقل برسد. با توجه به خاصیت بسیار عالی خاک دیاتومیت در جذب نانوذرات اکسید مس و جلوگیری از آزاد شدن آنها در محیط، استفاده از آن به همراه اکسید مس جهت کنترل باکتری، ضمن افزایش عملکرد، از اثرات سوء آن نیز جلوگیری می¬کند. اصلی¬ترین ترکیب موجود در این خاک سیلیس است که نوعی بازدارنده برای باکتری محسوب می¬شود. تحقیق حاضر به منظور بررسی امکان استفاده از دیاتومیت بهینه سازی شده با یون¬های مس در کنترل شیمیایی دو زیرگونه از باکتری Pectobacterium carotovorum در شرایط آزمایشگاهی انجام گردید. در ابتدا جهت دست¬یابی به بیشترین خلوص سیلیکا و همچنین باز¬تر کردن تخلخل آن به ترتیب از فرآیند¬های اسیدشویی و کلسیناسیون استفاده شد. نانو¬ذرات اکسید مس نیز از سولفات مس 5 آبه در آزمایشگاه سنتز شده و در خلل و فرج خاک جای داده شد. رشد جمعیت باکتریهای مورد نظر پس از تایید اولیه باکتری¬های مد¬نظر با روش¬های مولکولی، در محیط کشت مایع در حضور خاک دیاتومیت و اکسید مس مورد بررسی قرار گرفت. تغییرات جمعیت باکتری در ساعت¬های 0،9،24،48،72 اندازه¬گیری شد. نتایج حاصل شده نشان داد که نانوذرات اکسید مس بعد از 48 ساعت به تنهایی می¬تواند جمعیت باکتری Pcc را 8/0 درصد و باکتری Pco را 9 درصد کنترل کند اما با خاک دیاتومیت این مقادیر به ترتیب به 52/75 درصد و 55/90 درصد افزایش یافت. دو نانو¬ذره کبالت و نیکل نیز به نسبت 5 درصد وزنی خاک دیاتومیت مورد استفاده قرار گرفتند. در حضور نیترات کبالت 5 درصد باکتری Pcc به میزان 30/71 درصد و باکتری Pco 41/96 درصد کنترل شدند. در بررسی نتایج حاصل از نیترات نیکل نیز این نانو¬ذرات در نسبت 5درصد در مورد باکتری Pcc 91/59 درصد و در مورد باکتری Pco نیز به میزان 66/85 درصد باعث کاهش جمعیت باکتری گردید
Soft-rot disease is one of great economic issues in Iran and most parts of the world. Potato and carrot production, which are the most susceptible hosts to this disease has been reported significant damage each year. Many species of the genus Pectobacterium have been reported to cause soft rot. The two subspecies Pectobacterium carotovorum subsp. carotovorum (Pcc) and (Pco) Pectobacterium carotovorum subsp. odoriferum is one of the most important and common ones. Copper bactericidal properties have been used to control various pathogens since ancient times, but due to the adverse environmental effects of copper accumulation in plants, soil, water and food chains, efforts have been made to reduce consumption. At least reach Due to the excellent efforts of Diatomaceous earth in absorbing copper oxide nanoparticles and preventing their release into the environment, it is used to control bacteria while lowering the risk of heavy metal residues and also prevents its side effects. Also, the main compound in this soil is silica, which is a kind of inhibitor for bacteria. The current study was conducted to investigate the possibility of using diatomite optimized with copper ions in chemical control of two subspecies of Pectobacterium carotovorum in vitro. Initially, acid washing and calcination processes were used to achieve the highest purity of silica and porosity, respectively. Copper oxide nanoparticles were also synthesized from copper sulfate in the laboratory and coated on soil micro structures. The growth of the desired bacterial population was studied after initial confirmation using molecular methods in liquid culture medium in the presence of diatomaceous earth and copper oxide. Bacterial population changes were measured at 0,9,24,48,72 hours. The results showed that copper oxide after 48 hours alone can control Pcc 8% and Pco 9%, but with diatomaceous earth these percentages increased to 75/52% and 90/55%, respectively. Two nanoparticles of cobalt and nickel were also used in the proportion of 5% by weight of diatomaceous earth. Pcc bacterium was controlled in the presence of 5% cobalt nitrate by 71/30% and Pco bacterium by 96/41%. In the results of nickel nitrate, these nanoparticles in a ratio of 5% for Pcc showed 59/91% and for Pco showed 85/66% population control.
Investigation of possibility of using Diatomaceous earth optimized with Copper ions in chemical control of Pectobacterium spp. in vitro
