بررسی قرابت ژنتیکی گندم و خویشاوندان بومی آن از طریق الکتروفورز پروتئینهای مختلف ذخیرهای دانه

گونههای Aegilops و Triticum از خویشاوندان نزدیک گندم بوده و میصتوانند اهمیت بسزایی در اصلاح گندم داشته باشند .با توجه به اینکه این گونهصها در شرایط مختلف آب و هوایی و محیطی رشد میکنند منبع مناسبی برای ژنهای مقاومت به بیماریها، آفات، تنش های محیطی و ژنهای موثر در کیفیت نانوایی هستند .به منظور بررسی قرابت ژنتیکی ۱۶ گونه خویشاوند گندم از جنسهای Triticum و Aegilops از دو روشPAGE - SDSبا استخراج متوالی وPAGE - Acidاستفاده گردید .در هر دو سیستم ابتدا نوارهای پروتئینی با توجه به حرکت نسبی و مقایسه با نوارهای دیگر گونهها بسته به وجود یا عدم وجود نوار مقادیر صفر و یک دریافت کردند .سپس شباهت بین گونهصها با استفاده از معیار تطابق ساده محاسبه و با روش Centroid به دندروگرام تبدیل شد .بر اساس الکتروفورزPAGE (- SDSگلوتنینصهای با وزن مولکولی بالا(، گونههای مورد مطالعه در چهار گروه قرار گرفتند .گروه اول شامل گونههای T.dicoccoides ،T.durum ، T.araraticum و)T.aestivum گونهصهای پلیصپلوئید دارای ژنومA) ، گروه دوم شامل گونهصهای Ae.neglecta ، Ae.triuncialis ، Ae.kotschyi و) Ae. columnaris گونهصهای دارای ژنوم U جنسAegilops) ، گروه سوم شامل گونههایAe.tauschii ، Ae.speltoides ، Ae.biuncialis و Ae.cylindrica بالاخره گروه چهارم شامل گونههای T.urartu ، Ae.boeoticum ، Ae. umbellulata و Ae.crassa بود .در این سیستم دو گونه T.urartu و T.boeoticum در یک گروه قرار گرفتند .بدین ترتیب، گونههای حاوی ژنوم Aو U گروهبندی خوبی داشته و در گروههای مشخصی قرار گرفتند .بر اساس الکتروفورز پروتئینهای گلیادین نیز گونه ها در چهار گروه قرار گرفتند که با نتایج گلوتنین ها مطابقت دارد .همچنین بر اساس گلوتن) گلوتنین + گلیادین (گونههای مورد مطالعه در سه گروه واقع شدند و گونههای دارای ژنوم U یک گروه را تشکیل دادند .در روشPAGE - SDSبا استخراج متوالی بین تعداد نوار HMW و سطوح پلوئیدی رابطه روشنی مشاهده گردید .تعداد نوارهای HMW برای گونههای دیپلوئید ۱ تا ۲ عدد، برای گونههای تتراپلوئید ۳ تا ۴ عدد و برای گونههای هگزاپلوئید ۵ و بیشتربود .تجزیه تابع تشخیص برای گروههای دیپلوئید و تتراپلوئید بر اساس زیر واحدصهای HMW ، LMW ، گلوتنین وگلوتن معنیصدار بدست آمد ولی بر اساس گلیادین معنیصدار نبود .تجزیه تابع تشخیص برای مقایسه گونهصهای دارای ژنوم A با بقیه گونهصها نیز از نظر ویژگیهای مذکور معنیصدار بدست آمد.
The genera Triticum and Aegilops are wild relatives of wheat and they could be important in wheat breeding. They are found in a wide range of climatic regions and also grow on very different type of soils. Therefore, they are useful sources of genes for resistance to disease and insects, tolerance to environmental stresses and quality and quantity of grain protein. Genetic relations of 16 species belonging to Triticum and Aegilops genera were studied using SDS-PAGE with sequential extraction procedure and Acid-PAGE. At first, protein bands were scored as one and zero based on their existence or absence. Then, the data used to estimate genetic similarity between species by simple matching coefficient. Genetic similarities were converted to dendrograms by the centroid method. Genetic analysis using SDS-PAGE (HMW glutenin subunits) revealed four distinct groups of species. The first group contained T.durum, T.dicoccoides, T.aestivum and T.araraticum (polyploid Triticum species having A-genome), the second group included Ae.kotschy, Ae.triuncialis, Ae.neglecta and Ae.columnaris (Aegilops species with U-genome), the third group included Ae.speltoides, Ae.tauschii, Ae,biuncialis and Ae.cylindrica, and the fourth group contained T.boeoticum, T.urartu, Ae.umbellulata and Ae.crassa. In this condition T.urartu and T.boeoticum were placed in one group. The results of Acid-PAGE system were similar to the former analysis. S-genome and D-genome species could not be separated well from other species. One important result of this study was the correlation between the number of HMW glutenin subunits and ploidy level of species. Diploid, tetraploid and hexaploid species showed 1-2, 3-4 and 5 or more HMW subunits, respectively. Canonical discriminant analysis showed significant differences between tetraploid and diploid species and also between A-genome species and other species when used HMW, LMW, glutenin and gluten data.
