در این پروژه، شبیه سازی دینامیک مولکولی بر روی سلولز و سلولز آمینه انجام شد .آنالیزها نشان داد که ساختارهای طراحی شده پایداری لازم برای حمل دارو را دارا می باشند .به علاوه، ساختارها بر اساس آنالیزهای پیوند هیدروژنی، انرژی های الکتروستاتیک و وان دروالس دارای حساسیت به دما و pH نیز بودند .این آنالیزها نشان دادند که حضور گروه های آمینی در ساختار و پروتونه شدن آنها میزان برهم کنشهای بین مولکولی را کاهش می دهد .پس از آن، داروی پاکلیتاکسل از طریق داکینگ مولکولی در ساختارها بارگذاری شد و با شبیه سازی دینامیک مولکولی در دما و میزان پروتونه مختلف رفتار آن بررسی شد .آنالیزها نشان داد که افزایش دما و درصد پروتونه ساختار سبب کاهش برهم کنشهای میان دارو / حامل می گردد .در نهایت، جهت مقایسه نتایج شبیه سازی با آزمایشگاهی، سلولز و سلولز آمینه شده تهیه شدند و رهش داروی پاکلیتاکسل در مطالعه درون کشتگاهی در دو شرایط سلول های سرطانی و عادی بدن مورد بررسی قرار گرفت .نتایج آزمایشگاهی نشان داد که آمینه کردن سلولز سبب افزایش حساسیت به pH و دما سیستم حامل می شود و رهش دارو در شرایط سلول های سرطانی با کنترل رهش بهتری نسبت به سلول های عادی انجام می شود
In this project, molecular dynamics simulations were performed on cellulose and aminated cellulose. Analyzes showed that the designed structures had the required stability for drug delivery. In addition, structures based on hydrogen bonding analyzes, electrostatic energies and van der Waals were also sensitive to temperature and pH. These analyses showed that the presence of amine groups in their structure and protonation reduced the number of intermolecular interactions. Subsequently, paclitaxel was loaded into the structures via molecular docking and its behavior was studied by simulating molecular dynamics at different temperatures and protonation states. Analyzes showed that increasing the temperature and the protonation percentage of the structure reduced the drug/carrier interactions. Finally, to compare the results of the simulation with the experimental insights, cellulose, and aminated cellulose were prepared and the release of paclitaxel was evaluated in an in-vitro study in both normal and cancerous cell conditions. Experimental results showed that the amination of cellulose increases sensitivity to the pH and temperature of the carrier system, and drug release is achieved with better release control in cancer cell conditions than normal cells
