لوسمی میلوئیدی مزمن (CML) یک ناهنجاری تکثیر شونده میلوئیدی است که با توقف تمایز طبیعی، کاهش آپاپتوز و در نتیجه تکثیر سلولصهای خونی نابالغ و غیر عملکردی مشخص میصشود .با توجه به تأثیر رادیکالصهای آزاد در ایجاد سرطانصها و به دلیل عوارض جانبی مخرب و مقاومت دارویی در درمانصهای مرسوم، استفاده از ترکیبات جدید طبیعی با خواص آنتی اکسیدانی از رویکردصهای جدید درمانی میصباشد .تأثیر آنتی اکسیدانی ال-کارنیتین علاوه بر برداشت رادیکال-های آزاد از غشاء سلولی و کمک به اکسیداسیون اسیدهای چرب در جلوگیری از پیری سلولصها و تمایز سلولصها به ردهصهای مختلف بررسی شده است .هدف از این مطالعه بررسی اثر ال-کارنیتین بر بیان نشانگر سطح سلولی CD۳۴ به عنوان نشانگر مشترک در CML و پیش سازصهای مغز استخوان و همصچنین تأثیر آن بر میزان آپاپتوز و بیان ژن SUZ۱۲ میصباشد .در این مطالعه سلول-های تک هستهصای از مغز استخوان فمور رت جداسازی و کشت داده شدند .سلولصها تحت تیمار با غلظتصهای ۲/۰ و ۴/۰ میلی مولار ال-کارنیتین قرار گرفتند و سپس میزان بیان نشانگر سطح سلولی CD۳۴ به روش فلوسایتومتری، میزان آپاپتوز به روش Annexin V/PI و بیان ژن SUZ۱۲ به روشtime PCR - Realاندازه گیری شد .آنالیز آماری نتایج به کمک تست ANOVA با سطح اطمینان ۹۵ درصد ارزیابی گردید .به طور خلاصه مطالعه حاضر نشان داد در تیمار سلولصهای تک هستهصای مشتق شده از مغز استخوان با ۲/۰ میلی مولار ال-کارنیتین بیان نشانگر سطح سلولی CD۳۴ به میزان ۲۱/۰ درصد با ۰۵/۰>P افزایش یافته است .همصچنین در غلظت ۲/۰ میلی مولار ال-کارنیتین میزان آپاپتوز اولیه سلولصها ۳/۱۲ درصد با ۰۵/۰>P افزایش یافته است .در غلظت ۴/۰ میلی مولار ال-کارنیتین نیز بیان ژن SUZ۱۲ با ۰۱/۰>P به میزان ۷/۱ برابر گروه کنترل افزایش یافته است .نتایج حاصل از این تحقیق میصتواند به عنوان یک راهکار درمانی چند جانبه در روند بهبود بیماری CML نقش مؤثری داشته باشد
Chronic myeloid leukemia (CML) is a myeloid proliferative disorder characterized by blocking the natural differentiation and reducing apoptosis, as a result of the proliferation of immature and non-functional blood cells. Regarding the effect of free radicals on the development of cancers and because of adverse effects and drug resistance in conventional treatments, the use of new natural compounds with antioxidant properties is a new therapeutic approach. The anti-oxidant effect of L-carnitine has been investigated in addition to the removal of free radicals from the cell membrane and helping oxidation of fatty acids in preventing the aging of cells and differentiating cells into different classes. The aim of this study was to evaluate the effect of L-carnitine on the expression of CD34 marker as a common marker in CML and bone marrow precursors, as well as its effect on apoptosis and SUZ12 gene expression. In this study, bone marrow mono-nuclear cells (MNCs) of rat femur were isolated and cultured. The cells were treated with 0.2 and 0.4 mM L-carnitine concentrations, and then expression level of the CD34 cell line was measured by flow cytometry, apoptosis using Annexin V/PI method and SUZ12 gene expression by Real-time PCR. Statistical analysis was performed using ANOVA test with 95 confidence level. The results showed that in the treatment group of bone marrow-derived MNCs with 0.2 mM L-carnitine, cell surface marker expression of CD34 increased by 0.21 (P<0.05). Also, at the concentration of 0.2 mM L-carnitine, the early cell apoptosis increased by 12.3 (P<0.05). In the concentration of 0.4 mM L-carnitine, the SUZ12 gene expression increased by 1.7 fold when compared to the control group (P<0.01). The results of this study can be effective as a multidirectional therapeutic strategy for the improvement of CML
