سیستم های دارورسانی بر پایه نانو، پتانسیل بالایی در اثردهی بهتر داروهای ضدسرطان را فراهم نموده اند .ترکیب درمانی نیز یکی از شیوه های مناسب برای درمان بالینی سرطان محسوب می شود .از این رو، هدف از این پایان نامه، ارزیابی و طراحی نانودارورسان های مغناطیسی زیست سازگار و هوشمند برای انتقال همزمان داروهای ضدسرطان دوکسروبیسین (DOX) و متوتروکسات (MTX) به سلول های سرطانی سینه رده MCF۷ می باشد .از آنجایی که ساختار های شاخه دار می توانند دوز بالای دارویی با ویژگی های متفاوت را بارگیری نمایند، سه نوع نانودارورسان مغناطیسی بر پایه کیتوسان شاخه دار طراحی گردید .پس از سنتز و تائید زیست سازگاری نانودارورسان ها بر روی گلبول قرمز به کمک آنالیز همولایزیز، برهمکنش بین نانودارورسان با پروتئین های سرم خون انسان در سیستم گردش خون نیز به کمک شیوهPAGE - SDSمورد ارزیابی قرار گرفت .نتایج نشان داد که تعدادی پروتئین سرم خون به سطح نانوداروسان ها متصل شده و موجب شده است تا ویژگی گریز از سیستم ایمنی بدن مناسبی در سیستم شبیه سازی شده داشته باشد .مطالعات برون تنی نشان داد که دارورسانی دو دارویی اثردهی بالایی داشته و موجب کاهش اثرات جانبی شده است .علاوه بر این اثرات سمیت نانودارورسان بر روی رده سلولی MCF۷ مطالعه گردید و نتایج نشان داد که نانودارورسان ها مناسب برای سیستم های دارورسانی هستند .سایر آنالیزهای بیولوژیکی از جمله رنگ آمیزیDAPI ، آپتیک سلولی، چرخه سلولی و آپوپتوز نیز انجام شد .در نهایت اثرات جانبی نانودارروسان حاوی دارو و داروهای آزاد بر روی مدل حیوانی اجرا شد و نتایج نشان داد که ترکیب درمانی نانودارورسان های طراحی شده شیوه مناسبی برای درمان سرطان بوده که می تواند امکان استفاده در مطالعات بیشتر درون تنی را داشته باشد
Nanoparticulate drug delivery systems have the potential to improve the therapeutic efficacy of anticancer agents, and combination therapy is a promising strategy for clinical cancer treatment with synergistic effects. The purpose of this thesis is to evaluate smart, biocompatible, magnetic nanocarriers for the intracellular co-delivery of doxorubicin (DOX) and methotrexate (MTX) to the MCF7 cell line. Since dendritic architecture can load high doses of drugs with various properties, we designed three different type of dendritic chitosan based on magnetic nanoparticle as nanocarriers with potential in multi-drug delivery. After synthesis and confirmation of the nanocarriers biocompatibility with human red blood cells by hemolysis assay, SDS-PAGE assay was performed to simulate how the nanocarriers interact with proteins that exist in the blood circulation systems. The results show that several proteins in human blood plasma could attach to the nanocarriers surface and provide the nanocarriers with stealth properties that are suitable for drug delivery systems. An in-vitro study of cell viability showed that combination drug delivery has synergistic effects and reduces toxic side effects. Furthermore, the cytotoxicity assay of the nanocarriers to the MCF7 cell line indicated that desgined nanocarrier are suitable as anticancer drug nanocarrier. Dual-drug delivery with efficient anticancer performance was also confirmed by DAPI staining, cellular uptake, cell cycle, and apoptosis analysis, compared to free dual anticancer drugs. Finally, the side effects of the drug-loaded nanocarrier and free drugs were assessed using the mouse model and it was concluded that this combination therapy offers a promising approach to cancer treatment and can be used for further in-vivo applications
