هدف :از این مطالعه ارزیابی اثرات تقویت سیستم ایمنی باکتری کشته شده تسوکامورلا اینکونینسیس بر تغییرات تیتر آنتی بادی علیه واکسن نیوکاسل در بلدرچین ژاپنی می باشد .روش کار :بدین منظور جوجه ها از یک روزگی دوزهای مختلف باکتری را دریافت نمودند .در ۱ و ۷ روزگی به منظور بررسی تیتر مادری نیوکاسل خونگیری انجام شد .در روزهای ۱۰ و ۲۰ واکسیناسیون علیه بیماری نیوکاسل انجام) ب ۱و لاسوتا (شد .در روزهای۱۴ ،۲۱،۲۸،۳۵، ۴۲به منظور بررسی تیتر نیوکاسل، خونگیری انجام شد .در روزهای۷ ،۱۴ ،۲۱ ،۲۸ ، ۳۵ و ۴۲ به منظور تعیینFCR ، وزن کشی انجام شد .در روزهای۱۴ ، ۲۸ و ۴۲ به منظور بررسی تاثیر باکتری ایمنوژن بر ساختار بافتی روده و همچنین بررسی اثرات جانبی احتمالی، از بافت های مختلف نمونه گیری پاتولوژی انجام شد .در نهایت پس از جداسازی سرم از نمونه های خون، تیتر آنتی بادی ضد ویروس واکسن نیوکاسل با روش هماگلوتیناسیون و روش الایزا ارزیابی شد .از انجایی که اینترفرون آلفا، اینترفرون گاما و اینترلوکین ۴ در واکسیناسیون علیه بیماری نیوکاسل اهمیت ویژه ای دارند، فاکتورهای ذکر شده با استفاده از کیت های تجاری در گروه های مختلف اندازه گیری و مقایسه شد.نمونه های بافتی نیز در فرمالین فیکس و بعد از تهیه مقاطع و رنگ آمیزی هماتوکسیلین-ائوزین، مورد مطالعه هیستوپاتولوژی قرار گرفتند نتایج :در اندازه گیریIL - ۴وINF -در سرم گروه های مختلف آزمایشی در روزهای ۷ و ۴۲ تفاوت معنا دار دیده شد .در اندازه گیری IgAگروه دریافت کننده دوز بالا در روز ۴۲ تفاوت معنا داری نسبت به دو گروه دیگر داشت .باکترین تسوکامورلا اینکوننسیس باعث به وجود آمدن اختلاف آماری معنی دار در عیار آنتی بادی بر علیه نیوکاسل در سن۳۵ ،۲۸،۲۱،۱۴، ۷و ۴۲روزگی بین گروه کنترل با گروه های دریافت کننده باکترین و این گروه ها با یکدیگر شد .همچنین گروه های دریافت کننده باکترین در نمومه های بافتی گرفته شده از قسمت های مختلف روده با یکدیگر و با گروه کنترل در پارامترهای طول پرز، ضخامت پرز، عمق کریپت، تعداد کریپت و تعداد سلول های گابلت تفاوت معنا دار داشتند .در بررسی نتایج ضایعات هیستوپاتولوژیک بافت های مختلف در روز۱۴ در کبد، کلیه و قلب گروه دریافت کننده دوز بالا پرخونی خفیف و در کبد و کلیه هر ۳ گروه تغییر چربی خفیف دیده شد .در روز ۲۸ در کبد و کلیه گروه دریافت کننده دوز پایین پرخونی خفیف دیده شد .در روز ۴۲ هایپرپلازی به صورت خفیف در گروه دریافت کننده دوز بالا و به صورت متوسط در گروه دریافت کننده دوز پایین دیده شد . در گروه دریافت کننده دوز بالا در کبد و کلیه پرخونی متوسط و در قلب پرخونی خفیف دیده شد .در گروه دریافت کننده دوز پایین در کلیه پرخونی خفیف دیده شد . نتیجهگیری :با توجه به یافتهصهای ماکروسکوپی و سرولوژیک، میصتوان نتیجه گرفت که باکتری کشته شده تسوکامورلا اینکوننسیس باعث تغییرات تیتر آنتی بادی علیه واکسن نیوکاسل در بلدرچین ژاپنی، بدون به وجود آوردن ضایعات پاتولوژیک خاص شد .باکتری کشته شده تسوکامورلا اینکونینسیس می تواند بدون ایجاد عوارض جانبی در بافت های حیاتی، موجب بهبود ساختار هیستومورفولوژیک روده کوچک به ویژه در فاکتورهای طول پرزها، ضخامت پرزها و عمق کریپت ها و بیشتر در قسمت ایلئوم و در دوز بالاتر در جوجه بلدرچین ژاپنی شود
Objective: The aim of this study was to evaluate the effects of immunocompromised Tsukamurella inchonensis bacteria on antibody titer changes against Newcastle disease vaccine in Japanese quail. Methods: For this purpose, chicks received different doses of bacteria from a single day. At 1 and 7 days, blood samples were collected from Newcastle's native Newcastle disease. On 10 and 20 days, vaccination against Newcastle disease was performed (B1 and Lasotta). Blood sampling was performed on Newcastle disease in the days 14,21,28,35,42. Weighing was done on days 7, 14, 21, 28, 35 and 42 to determine FCR. On 14, 28 and 42 days from different tissues of pathologic sampling, an immunogenic bacterial effect on the histological structure of the intestine was investigated as well as a possible side effect study. Ultimately, after serum isolation from the blood samples, the Newcastle disease vaccine anti-virus antibody titer was assessed by haemagglutination and ELISA. Since IFN-, interferon-gamma, and IL-4 have an important role in vaccination against Newcastle disease, the factors mentioned were measured and compared by using commercial kits in different groups. Tissue specimens were also found in formalin fix and After staining and staining of hematoxylin-eosin, they were examined histopathological. Results: There was a significant difference in the measurement of IL-4 and INF- in serum of experimental groups on days 7 and 42. In the measurement of IgA, the high dose recipient group had significant differences in day 42 with respect to the other two groups. The bacillus Tsukamurella inchonensis caused a statistically significant difference in antibody titer against Newcastle disease at the age of 35,28,21,14,7 and at 42 days of age between the control group and the most frequent recipient groups. Also, the bacterial receptor groups were significantly different in the tissue stomachs taken from different parts of the intestine with each other and with the control group in parameters of lamellar length, lamellar thickness, crypt depth, crypt number and number of goblet cells. In histopathological lesions of different tissues on the 14th day in the liver, the kidneys and heart of the high-dose group showed mild prevalence, and in the liver and kidneys of all three groups, moderate lipid changes were observed. On the 28th day in the liver and kidneys, a low dose of low-dose transfusions was observed. On day 42, hyperplasia was mild in the high dose group and moderately in the low dose group. In the high dose group in the liver and in the kidney, moderate hyperemia and mild hyperthyroidism were observed. In the low-dose group, there was a mild hyperemia in kidney. Conclusion: According to macroscopic and serologic findings, it can be concluded that the killed bacterium Tsukamurella inchonensis caused changes in the antibody titer against Newcastle disease vaccine in Japanese quail without causing specific pathological lesions. The deadly bacteria Tsukamurella inchonensis can improve the histomorphological structure of the small intestine, especially in lung length factors, the thickness of the lice and the depth of the crypts, and more in the ileum and at a higher dose in Japanese quail chicks
