پیش زمینه- کورکومین یک عامل ضد التهابی و آنتی اکسیدان قوی است .به دلیل اب گریز بودن کورکومین این ماده روی نانوذرات زئولیت با خاصیت بارگذاری شد .در این مطالعه یک سیستم دارورسان زیست سازگار متشکل از نانوزئولیت بارگیری شده با کورکومین تهیه و برای بهبود زخم بر روی پوست مورد استفاده قرار گرفت .روش کار- نانوکامپوزیت زئولیت-کورکومین/ ژلاتین با روش سل- ژل تهیه شده با استفاده از روشهای XRD و SEM شناسایی شد .میزان بارگیری کورکومین و رهایش ان و تست سمیت سلولی (MTT assay) نانوکامپوزیت تهیه شده انجام گرفت .تحت بیهوشی عمومی زخم تمام ضخامت به ابعاد۱.۵ ۱.۵cmدر ناحیه پشتی رتها ایجاد شد .حیوانات بصورت تصادفی و مساوی به پنج گروه پنج تایی: گروه) ۱کنترل، نرمال سالین (گروه) ۲ ژلاتین (و گروه ۳ کورکومین) کورکومین (گروه چهار زئولیت) زئولیت (و گروه پنج)نانوکامپوزیت زئولیت-کورکومین/ژلاتین (تقسیم شدند .درمانهای هر گروه بصورت موضعی روزانه یکبار تا ۱۴ روز روی زخم قرار میگرفتند .در روزهای صفر،،۳ ،۶ ،۱۲ ۹ ، و ۱۴ از محل زخمها عکس برداری شده و انقباض زخم انها محاسبه میشد .در روز ۱۴ حیوانات به روش انسانی قربانی شده و نمونههای پوستی جهت ارزیابی هیستوپاتولوژیک) رنگ امیزی هماتوکسیلین-ائوزین (برداشته شدند .نتایج- در این مطالعه نانوکامپوزیت زئولیت-کورکومین/ژلاتین با موفقیت تهیه شد .نتایج تست سمیت نانوکامپوزیت نشان داد که تا غلظت ml/mg ۱ سمیت ندارد .در گروه درمان شده با نانوکامپوزیت انقباض چشمگیر زخم و تسریع زمان التیام مشهود بود .(p<۰.۰۵) بعلاوه گروه درمان شده با نانوکامپوزیت در مقایسه با بقیه گروههای مطالعه سنتز بیشتر کلاژن و تسریع اپیتلیزاسیون و کمترین میزان عفونت در محل التیام را نشان داد .نتیجه گیری و کاربرد بالینی باتوجه به نتایج مطالعه حاضر میتوان چنین نتیجه گیری کردکه نانوکامپوزیت زئولیت / ژلاتین با سازگاری بافتی بالا میتواند برای تحویل موضعی کورکومین در محل زخم مفید باشد .
1.5 cm was created on dorsal area of 25 rats. The animals were equally and randomly divided into five groups of 5 each i.e. group I (0.9 sodium chloride), group II (gelatin treated) and group III (curcumin treated) GroupIV (zeolite treated) and group V (nanocomposite treated). The solutions and the formulation were applied topically on the wound once daily for 14 days. Photograph of each wound was taken on days 0,3,6,9,12 and 14 post wound creation. The area of wound was determined planimetrically. At 14 days, animals euthanized and skin samples were taken to histopathologicl evaluation (H&E staining). Results- In this work, we successfully prepared zeolitecurcumin/gelatin nanocomposite. The MTT test results indicate nanocomposite is safe up to 1mg/ml of nanocomposite. Nanocomposite treated group exhibited enchantment of wound closure and accelerate wound healing time (p<0.05). Furthermore, nanocomposite treated group showed complete re-epithelialization, more collagen synthesis and the least amount of secondary infection among the study groups. Conclusion and clinical relevance- In conclusion, based on the results of the present study, we concluded that Zeolite /gelatin nanocomposite can be useful for local delivery of curcumin with high viability in woundsBackground- Curcumin is believed to be a potent anti-inflammatory and antioxidant agent.
