Medical plants are an important source of secondary metabolites which is widely used in the field of drug development and treatment of disease. Tissue culture and micropropagation are also important tools to increase the production of effective materials under controlled conditions. Acinos graveolens of the family Lamiacae is a well-known medicinal plant with strong antimicrobial activity. In the present study, in vitro shoot culture of Acinos graveolens was established using nodal segments. We investigated the effect of different concentrations of sucrose on the in vitro organogenesis of A. graveolens by using Murashige and Skoog culture medium of different concentrations of sucrose (10, 20, 30, 40, 50gr). The highest dry and fresh wiegth were obtained by nodal segments cultured in MS medium with 30 gr sucrose. The highest mean flower and leaf number were achieved by nodal segments cultured in MS medium with 40gr sucrose. The maximum shoot leangh, the highest mean shoot number and SFC index were achieved by nodal segments cultured in 30gr sucrose. The best of medium for regeneration of this plant was medium with 20gr sucrose. The maximum root leangh, the highest mean root number and RFC index were achieved by nodal segments cultured in 40gr sucrose. The qualitative and quantitative changes of volatile compounds were observed in response to the different concentrations of sucrose. The maximum amount of pigments were achived by explants on MS medium with 20gr sucrose. Nevertheles, the highest amount of phenolics and flavonoids were observed on medium with sucrose 40 and 50gr. The results of this study showed that the most appropriate culture medium for production of some secondary metabilites such as phenolic and flavonoids is culture medium with high sucrose
