Turmeric (Curcuma longa) is reported to have hepatoprotective, anti-inflammatory, anticarcinogenic, and antimicrobial effect. In this study thirty male Wistar rats weighing 150-200g was divided into 5 groups of 6 rats. Rats in groups A ,B and C received 200 mg/kg, 400 mg/kg and 600 mg/kg body weight of the ethanolic extract of C. longa while rats in group D and E received Dimethyl sulfoxide (DMSO) and distilled water respectively. The regimen was administered orally by gavage once daily for 60 days and the rats were observed for signs of illness and/or death. At the end of the dosing period, the rats were sacrificed, blood samples were collected and analyzed. The liver and kidney were also harvested for gross and histopathological examination. The result showed that the LD50 was above 5000mg/kg. Turmeric induced anemia as it significantly (p<0.05) reduced Packed Cell Volume (PCV) and total red blood cell at 400 mg/kg and 600 mg/kg when compared with the control groups. There was also anemia resulting from a significantly (p<0.05) reduced packed cell volume and total red blood cell in the group exposed to DMSO. The study also showed that turmeric caused an increase in total leucocyte count (leukocytosis) with apparent increase in absolute neutriphil count (neutrophilia). There was a significant increase in serum AST and decrease in serum ALP activities in between the C.longa treated groups and the control group. There was also a significant (p<0.05) increase in percentage body weight at 600mg/kg but no significant difference (p>0.05) in the concentrations of total protein, globulin, albumin and serum electrolyte and the relative organ weight of the liver and kidney of groups A, B, and C when compared with the control group. The total protein and globulin concentrations were significantly (p<0.05) lower when compared with the DMSO group. DMSO also caused a significant decrease (p<0.05) in relative organ weight of the kidney. Histological examination of the liver and kidney showed some common changes with no definite pattern in all treated groups. The severity did not relate to the dose of turmeric or DMSO. Conclusively, the study showed that subchronic exposure to turmeric is safe at a low dose but prolonged use at high dose may cause anemia.
