هدف از مطالعه حاضر بررسی چند شکلی دو ژن کاندیدای لپتین) شامل دو جایگاه اگزون ‮‭۲‬ و اینترون ‮‭۲)‬ و گیرنده هورمون رشد) قسمتی از پروموتور (و تأثیر آنها بر صفات کیفی منی و برآورد پارامترهای ژنتیکی گاوهای نر هلشتاین بود .به این منظور مجموعا ‮‭۶۷‬ گاو نر از دو ایستگاه اصلاح نژاد شمالغرب کشور ‮‭(۴۱‬ گاو نر (و مرکز اصلاح-نژاد و بهبود تولیدات دامی کشور ‮‭(۲۶‬ گاو نر (انتخاب شد که در بین سالهای ‮‭۱۳۸۲ - ۱۳۹۲‬ اسپرم‌صگیری شده بودند .چهار صفت مورد مطالعه‌صی مربوط به کیفیت منی شامل حجم انزال) سی‌صسی(، جمعیت اسپرم) میلیون در سی‌صسی(، درصد اسپرم زنده قبل از انجماد) درصد (و درصد اسپرم زنده بعد از ذوب) درصد (بود .استخراج ‮‭DNA‬ از اسپرم با استفاده از روش نمکی با کمی تغییرات انجام شد، سپس ارزیابی چندشکلی در دو جایگاه ژن لپتین) اگزون ‮‭۲‬ و اینترون ‮‭۲)‬ به روش‮‭RFLP -PCR‬و در ژن گیرنده هورمون رشد) پروموتور (با روش ریزصماهواره انجام شد .پارامترهای ژنتیکی و همبستگی‌صهای ژنتیکی و محیطی همراه اثر ژنهای کاندیدا با روش بیزی برآورد گردید .نتایج نشان داد که تمام جایگاههای مورد بررسی چند شکل بوده‌صاند .بررسی‌صهای آنالیز واریانس نشان داد که ارتباط معنی‌صداری (>‮‭(۰۱/۰P‬ بین جایگاههای مورد نظر با صفات کیفی منی وجود دارد .در هر جایگاه‌ص‍ دسته بندی شده اختصاصی، ژنوتیپ ‮‭AA‬ جایگاه اگزون ‮‭۲‬ ژن لپتین برای صفات حجم منی و درصد زنده قبل از انجماد، و ژنوتیپ ‮‭TT‬ این جایگاه برای صفات جمعیت اسپرم و درصد زنده مانی بعد از انجماد، ژنوتیپهای مطلوب بودند .نتایج نشان داد در جایگاه اینترون ‮‭۲‬ این ژن، ژنوتیپ‮‭AA‬ ، و در جایگاه پروموتور ژن گیرنده هورمون رشد نیز ژنوتیپ‮‭۱۷/ TG۱۷ ,TG‬ ژنوتیپ‌صهای مطلوب برای بیشتر صفات و کیفیت منی، مخصوصا پتانسیل منی برای عمل انجماد مؤثر بوده است .تخمین وراثت‌صپذیری توسط آنالیز چندصصفته و تک‌صصفته با مدل حیوانی تکرارپذیری برای صفات حجم منی، جمعیت اسپرم، درصد اسپرم زنده قبل انجماد و بعد ذوب بترتیب‮‭۱۱/۰‬ ،‮‭۱۹/۰‬، ‮‭۲۴/۰‬ ، ‮‭۲۳/۰‬ و‮‭۲۲/۰‬ ،‮‭۱۴/۰‬ ،‮‭۲۰/۰‬ ، ‮‭۱۰/۰‬ بدست آمد .بیشترین همبستگی‌صهای ژنتیکی مربوط به صفات درصد اسپرم زنده قبل انجماد با درصد اسپرم زنده بعد ذوب، به مقدار ‮‭۹۱۲/۰ ۰۳۱/۱۱‬و کمترین آن مربوط به صفات حجم منی با غلظت اسپرم، به مقدار ‮‭۲۸۰/۰ ۸۰/۸۶‬ - بود

The purpose of this study was to estimate the genetic parameters for some of the semen quality traits with evaluation the polymorphism of candidate genes including Leptin (exon 2 and intron 2) and Growth hormone receptor (promoter) genes and their effects on semen quality traits of Holstein bulls. For this purpose data information form two breeding stations including the Northwest Center of Animal Breeding (41bulls) and the Country Breeding Center of Iran (26 bulls) were used, totally information of 67 bulls, were used, That were sampled between 1382-1392 of years. Four sperm qualitative traits, including volume of ejaculate (cc), sperm concentration (million/cc), and the percentage of live sperm before freezing ( ) and percentage of live sperm after thawing ( ) were considered. The polymorphism in the Leptin gene evaluated by PCR-RFLP and the variation in the Growth hormone receptor gene were evaluated by PCR-SSR. Genetic parameters with candidate gen effects were estimated by Bayesian method. The result of this study showed that all loci were polymorphism. Analysis of variance revealed signi?cant association of investigating loci on semen quality (p<0.01). When loci classified into their individual genotype, a significantly effect of AA genotype of exon-leptin on volum and percentage of live sperm before freezing and also TT genotype of exone-leptin significantly affect sperm concentration and percentage of live sperm after thawing. results showed, AA genotype of intron-leptin gene and TG17/17 was favorable for most investigated semen charatristics and may affect on semen quality and specially on the potential of semen for freezing. Estimation of heritability by multivariate and one trait with repeatability animal model for semen volume, sperm concentration, live sperm before freezing and live sperm after thawing were obtained 0.11, 0.19, 0.24, 0.23 and 0.22, 0.14, 0.20, 0.10 respectively. The high Genetic correlation between percentage of live sperm before freezing and percentage of live sperm after thawing showed 0.9111.03 and low Genetic correlation between volum , sperm concentration were observed -0.2 86.80
