در سال های اخیر، با شناسایی و اثبات تاثیرات مثبت میکروب های پروبیوتیک بر روی سلامت انسان ها، استفاده از پروبیوتیک‌ها به طور چشمگیری رو به افزایش است .از عمده میکروارگانیسم‌های پروبیوتیک، باکتری‌ها هستند که در این میان لاکتوباسیلوس‌ها از معروف‌ترین آن‌ها می‌باشند .لاکتوباسیلوس‌ها باکتری‌های گرم مثبت، میله‌ای، بدون اسپور و اکثرا بی‌هوازی اختیاری یا میکروآئروفیل هستند که در گروه" عموما سالم شناخته شده "یا اصطلاحا ‮‭GRAS‬ قرار دارند .آن‌ها جزو فلور طبیعی دستگاه گوارش بوده و تاثیرات مفید زیادی بر روی سلامت انسان می‌گذارند .از مهم‌ترین اثرات مفید لاکتوباسیلوس‌ها می‌توان به جلوگیری از سرطان‌ها، تحریک سیستم ایمنی، تولید ویتامین‌ها، کمک به کاهش کلسترول خون، جلوگیری از کلونیزاسیون باکتری‌های بیماری‌زا در روده و غیره اشاره کرد .لاکتوباسیلوس‌ها باکتری‌های پرنیازی هستند که به منظور کشت این باکتری‌ها عمدتا از محیط کشت ‮‭MRS‬ ، ‮‭Skim Milk‬ و ... استفاده می‌شود .هزینه‌ی ساخت این محیطهای کشت به دلیل پر نیاز بودن باکتری بسیار بالا می‌باشد .بدین منظور طراحی و ساخت یک محیط کشت ارزان قیمت می‌تواند کمک زیادی در زمینه کشت لاکتوباسیلوس‌ها درمقیاس بالا در صنایع غذایی و دارویی بکند .هدف از این تحقیق، معرفی یک محیط کشت بهینه ارزان قیمت به منظور کشت لاکتوباسیلوس کازئی‮‭UT۱‬ ، با هدف دسترسی به توده سلولی بالا نسبت به محیط های مرسوم آزمایشگاهی است .به منظور طراحی محیط کشت بهینه برای لاکتوباسیلوس‌ها، از طراحی آزمایش یا‮‭DOE‬ استفاده می‌کنیم .یکی از قدیمی‌ترین روش‌های ‮‭DOE‬ روش تک متغیره می‌باشد که به دلیل هزینه‌ی بالا و نیاز به زمان زیاد کارایی کمی دارد .در این پژوهش از میان روشهای طراحی آزمایش، از روش تاگوچی به دلیل هزینه‌ی پایین، سرعت بالا و در عین حال دقت بالای این روش استفاده شد .ابتدا با استفاده از روش کاملا تصادفی، منابع کربن و نیتروژن بهینه انتخاب شد .سپس با استفاده از روش طرح آزمایش تاگوچی، میزان اثر فاکتورهای مختلف بر روی رشد باکتری ‮‭L.casei UT۱‬ بررسی شد .محیط کشت بهینه به دست آمده حاوی ترکیب ملاس و آب پنیر با نسبت برابر‮‭(MW)‬ ، عصاره مخمر، استات سدیم، سولفات منیزیم و سولفات منگنز به ترتیب با مقادیر ‮‭۸‬ ، ‮‭۴‬ ، ‮‭۱‬ ، ‮‭۰۴/۰‬ و ‮‭۰۰۵/۰‬ بود که میزان رشد باکتری را به میزان ‮‭۸۹/۱۱۴‬ درصد نسبت به محیط کشت ‮‭MRS‬ افزایش داد .این محیط کشت طراحی شده، به دلیل افزایش روز افزون در مصرف محصولات غذایی پروبیوتیک و نیاز به کشت پروبیوتیک ها در مقیاس زیاد، می‌تواند به تولید این محصولات غذایی کمک زیادی کند

scale probiotic fermentation.-designed culture medium can greatly contribute to the production of probiotic food products due to the increasing use of probiotic food products and the need for large-called GRAS. They are part of the natural flora of the gastrointestinal tract and have many beneficial effects on human health. The most important effects of Lactobacillus include preventing cancers, stimulating the immune system, producing vitamins, helping to lower blood cholesterol, preventing colonization of pathogenic bacteria in the gut, and so on. Lactobacillus are fastidious bacteria and MRS, Skim Milk etc , are mainly used for their cultivation. The cost of producing these media is very high due to the bacteria complex nutrition. To this purpose, designing a modified and inexpensive culture medium can greatly help in cultivating Lactobacillus at large scales in the food and pharmaceutical industries. The purpose of this study was to introduce an economical culture medium for the cultivation of Lactobacillus casei UT1, to access high cell mass compared to conventional laboratory media. To design the optimum culture medium for Lactobacillus, we used design of experiments (DOE). One of the oldest methods for DOE is the univariate method, which is less efficient because of it is expensive and time consuming. In this study, the Taguchi method was used for testing because of its low cost, high speed, and high accuracy. First, optimum carbon and nitrogen sources were selected using a completely randomized factorial method. Then, the effect of different factors on the growth of L.casei UT1 was evaluated using Taguchi's experimental design method. The optimum culture medium containing molasses and whey with equal ratio (MW), yeast extract, sodium acetate, magnesium sulfate, and manganese sulfate was 8 , 4 , 1 , 0.04 and 0.005, respectively. It was increased bacterial growth rate by 114.4 compared to the MRS medium.
