سرطان کولورکتال بالاترین میزان مرگ و میر را در سراسر دنیا دارد و دلیل اصلی مرگ ناشی از سرطان در کشورهای اروپایی محسوب می‌شود. شناخت ژن‌ها و مسیرهای درگیر در سرطان کولورکتال می‌تواند در تشخیص زود هنگام این بیماری و در نتیجه کاهش میزان مرگ و میر مؤثر باشد؛ بنابراین بررسی‌های بیشتر مولکولی برای شناسایی بیومارکرهای جدید برای پیشگیری، تشخیص زودهنگام، طبقه بندی بهتر بیماری و انتخاب روش درمانی ضروری است. از بیومولکول‌هایی که امروزه بیشتر مورد توجه محققان قرارگرفته، می‌توان به RNA‌های غیر کد کننده طولانی (Long non coding RNA) اشاره کرد که مطالعه‌های جدید نشان داده‌اند که نقش مهمی در پیشرفت سرطان و مقاومت به درمان دارند. LncRNA-VLDLR به طور قابل توجهی در سلول‌های بدخیم مشاهده شده است. جامعه آماری تحقیق را بیماران مبتلا به سرطان کولورکتال که از اردیبهشت تا اسفند 1400 در بیمارستان شهيد محلاتي شهر تبریز بستری بود تشکیل می‌دهد. در این مطالعه میزان بیان ژن LncRNA-VLDLR در سلول‌های سرطانی کولورکتال و سلول‌های سالم مقایسه شد و نتایج این تحقیق نشان داد که میانگین بیان ژن LNCRNA- VLDLR در مردان بیشتر از زنان و در سن بالای 55 سال نسبت به سن زیر 55 سال بیشتر بیان شده و از نظر آماری معنی‌دار می‌باشد. با در نظر گرفتن مرحله‌ی بالینی تومور، نمونه‌هایی که در Stage III قرار داشتند و تومورهایی که در مرحله‌ی بالینی یک و دو قرار داشتند علاوه بر افزایش بیان، از نظر آماری معنادار بودند. نتایج نشان داد که رابطه‌ی معناداری میان افزایش بیان Lnc-VLDLR و تهاجم سلولی مشاهده شد. همچنین طبق نتایج مطالعه‌ی انجام شده، بیان ژن LNCRNA- VLDLR در بافت توموری سرطان کولورکتال نسبت به بافت سالم حاشیه‌ای به طور معنی‌داری افزایش پیدا کرده است؛ بنابراین می‌توان با مطالعات گسترده و حجم نمونه بیشتر ژن LNCRNA- VLDLR به عنوان یکی از بیومارکر‌های تشخیصی یا پیش بینی کننده برای سرطان کولورکتال در آینده استفاده کرد.

Abstract:Colorectal cancer has the highest mortality rate worldwide and is the leading cause of cancer death in European countries. Identification of genes and pathways involved in colorectal cancer can be effective in early diagnosis of the disease and thus reduce mortality; Therefore, further molecular studies are necessary to identify new biomarkers for prevention, early diagnosis, better classification of the disease, and treatment options. One of the biomolecules that has attracted the most attention of researchers today is long non-coding RNAs, which new studies have shown play an important role in cancer progression and resistance to treatment. LNCRNA-VLDLR has been significantly observed in malignant cells. The statistical population of the study consists of patients with colorectal cancer who will be hospitalized in Mahalati Tabriz Hospital in Tabriz from May to March 2014. In this study, the expression of LNCRNA-VLDLR gene in colorectal cancer cells and healthy cells was compared. And is statistically significant. Considering the clinical stage of the tumor, the samples that were in Stage III and the tumors that were in clinical stage one and two, in addition to LNCreasing the expression, were statistically significant. The results showed that a significant relationship was observed between LNCreased LNC-VLDLR expression and cell invasion. Also, according to the results of the study, the expression of LNCRNA-VLDLR gene in colorectal cancer tumor tissue has LNCreased significantly compared to marginal healthy tissue. Therefore, with extensive studies and larger sample sizes, the LNCRNA-VLDLR gene can be used as one of the diagnostic or predictive biomarkers for colorectal cancer in the future.
