Improvement of wheat (Triticum aestivum L.) by biotechnological approaches is currently limited by a lack of efficient and reliable tissue culture methodology. In this research, immature embryos of seven Iranian bread wheat cultivars viz. Alvand, Azadi, Atrak, Atilla4Y, Atilla50Y, Tajan and Falat were used to evaluate their ability of callus induction ,green spot formation and shooting. A split - split plot based on completley randomized design with 4 replications were adapted in which Factor A included of the cultivars, factor B consisted of four embryogenic callus induction medium, and factor C constituted of three regeneration medium. Analysis of variance showed a significant diference between cultivars for all the invitro traits studied except callus induction after 30 days from culture. Significant difference was found for factor B(callus induction medium)for all the traits studied. The interaction of cultivar* callus induction medium was significant for callus induction after 15 as well as 30 days from culture, volume of callus after 15 days from culture, green spot after 7, 21, 28 days from subculture, number of green spot per callus after 14 day from culture, percentage of shoot formation, and regeneration frequency. Factor C(regeneration medium) and its interaction with factors A and B was significant with respect to green spot after 7, 21, 28 days from subculture, number of green spot per callus after 14 days from culture, percentage of shoot formation, regeneration frequency.
