.2 oC in growth chamber. This study was carried out factorial experiment based on completely randomized design with four replications. After 4 weeks of culture, number of lateral shoots and node, shoot length, root and callus formation percentages were measured. Analysis of variance showed that number of lateral shoots and node, shoots length, root and callus formation percentages were influenced by the factors. Maximum number of lateral shoots was observed in media including 50 mg.l-1Nicotinic acid, 50 mg.l-1 Pyridoxine- HCl, 10 mg.l-1 Thiamine-HCl and 4 mg.l-1 BAP. Main shoot length was not significantly affected by different concentrations of MS vitamins. Thiamine-HCl concentrations had an important role in number of main shoot nodes in media as observed for high concentrations of BAP. Maximum percentage of root and callus formation was obtained in media without BAP and with different concentrations of MS vitamins 2 oC. Analysis of variance revealed significant differences among treatments for initiation and formation of microtuber, length, diameter, weight and rate of initiation. Sucrose had the highest effect on microtuberization process when compared with mannitol. The mannitol in the media resulted in a decrease in microtuberization. Increasing of sucrose concentrations were efficiently improved in vitro microtuber production without negative side effects. Therefore, the use of high levels of sucrose was useful on in vitro microtuberization
