The wider applications of nanoparticles (NPs) have evoked a world-wide concern due to their possible risk of toxicity in humans and other organisms. Iron oxide nanoparticles (IONs) can easily accumulate in different tissues and induce toxicity through induction of oxidative stress, mitochondrial changes, membrane disruption, DNA damage, cell cycle arrest, apoptosis, cytoskeletal disruption and gene expression alteration. Cells respond to various environmental stresses including high and low temperature, chemicals, physical damages and radiations via different molecular mechanisms. Heat shock proteins are among the main cellular system for adaptation with different stressful conditions. Despite the increasing relevance of IONs and their applications in biosystems there are no reports covering small heat shock proteins (sHSPs) expression in chicken brain and in response to NPs. Therefore, the current study conducted to evaluate the expression of HSPB1 and HSPB5 in the chickens embryo brain exposed to the IONs. Two hundred-forty Ross 308 embryonated eggs were randomly divided in eight groups with three repeats. Negative control group received nothings. The positive control group received 0.3 ml of normal saline. Groups 3-5 were administrated 0.3 ml of maghemite suspension with concentrations of 100 ppm, 250 ppm and500 ppm, respectively. Groups 6-8 received 0.3 ml of magnetite suspension with concentrations of 100 ppm, 250 ppm and500 ppm, respectively. Eggs were inoculated by In ovo method at day one of embryonic period and incubated at 37.5 C with 60 humidity. After morphological evaluation at day 20, the brains were harvested in sterile condition to evaluate the expression of HSPB1 and HSPB5. The expression level of target genes were analyzed using qPCR with SYBR Green I and 2-Ct method. The results showed that HSPB1 and HSPB5 are expressed in the chicken embryo brain and upregulate in response to the different forms of IONs dose dependently. Beside other biological functions, sHSPs are also involved in cell response to metal oxide NPs
