درختان صنوبر به جنس Populus و خانواده Salicaceae تعلق دارند و بومی نواحی مختلفی در نیم کره شمالی میصباشند .این درختان در استان آذربایجان صغربی بهص طور گسترده در جنگلصداری یا همراه با سیستم کشاورزی استفاده می-شوند .بیماریهای زیادی توسط میکروارگانیسمهای مختلف در ریشه، تنه و شاخسار درختان صنوبر ممکن است ایجاد شود .که از مهمترین آنصها میتوان به شانکر تنه و سرخشکیدگی اشاره کرد که به عنوان یک تهدید بالقوه در مناطق کشت و پرورش درختان صنوبر در دنیا به حساب میصآید و در نهایت به زوال و مرگ درختان صنوبر میصانجامد .گونهصهای متعددی از قارچصهای جنسCryptosphaeria ،Cytospora ،Necteria ، Leucostoma و Botryosphaeria در ایجاد این نوع بیماریصها دخیل هستند .هدف تحقیق حاضر شناسایی عوامل قارچی همراه با بیماری شانکر تنه و سرخشکیدگی درختان صنوبر در استان آذربایجان غربی با استفاده از دادهصهای مورفولوژیکی و مولکولی میصباشد .برای این منظور در طی تابستان و پاییز سال۱۳۹۴ ، نمونهصبرداری از درختان صنوبر و بید دارای علائم شانکر صورت گرفته و تعداد ۱۱۰ عدد نمونه مورد بررسی قرار گرفت .جداسازی و خالصصسازی قارچ عامل بیماری به روش تک کنیدی بر روی محیط کشت آب-آگارA) - (Wانجام شد و تعداد ۱۷۳ جدایه قارچی از نمونهصهای دارای علائم شانکر جداسازی گردید .هویت گونهصها با بررسیصهای ریختصشناختی و مراجعه به منابع و توصیفصهای معتبر شناسایی و در نهایت با استفاده از توالی دادهصهای ناحیهrDNA - ITSمورد ارزیابی قرار گرفت .به منظور بررسی بیماریصزایی جدایهصهای قارچی، آزمون بیماریصزایی هر یک از جدایهصها در شرایط گلخانهصای و بر روی دو رقم Populus nigra و P. alba انجام گرفت .نتایج این بررسیصها نشان داد که گونهصهای قارچی همراه با بیماری شانکر تنه و سرخشکیدگی درختان صنوبر و بید در این منطقه شاملCryptosphaeria pullmanensis ، Cytospora chrysosperma و .Cytospora sp میصشوند .در بین گونهصهای قارچی جداسازی شده C. pullmanensis با فراوانی ۸۴ درصد به عنوان گونه قارچی غالب معرفی می-گردد .این گونه فقط از روی درختان صنوبر جداسازی گردید .نتایج حاصل از مایهصزنی، بیماریصزا بودن همه جدایهصهای قارچی را تأیید کرد .جدایه .Cytospora sp روی P. nigra بیشترین طول زخم با اندازه cm ۷۵/۲۲ ایجاد نموده و روی P. alba با cm ۶ دارای کمترین مقدار بود و به نظر میصرسد که حساسیت P. nigra نسبت به گونهصهای سیتوسپورا به مراتب بیشتر از p. alba باشد .اما میانگین طول زخم روی چوب توسط Cryptosphaeria pullmanensis روی P. alba به نسبت P. nigra بیشتر بود، در نتیجه به نظر میصرسد که P. alba نسبت به P. nigra حساسصتر باشد

Poplar trees (Populus spp.) belong to the Salicaceae family and are native to various regions in the northern hemisphere. These trees in West Azerbaijan province are extensively used in forestry or integrated with agricultural systems. Several diseases may be created by various microorganisms in the roots, trunk and branches of poplar trees. Trunk canker and dieback diseases are among the most important diseases, on poplars all over the world. These diseases are considered as potential threats on poplar trees and are involved in decline or slow death of poplar trees. Various species from different fungal genera including Cryptosphaeria, Cytospora, Nectria, Leucostoma and Botryosphaeria are involved in occurrence of the disease. The purpose of the current study was to identify fungal agents associated with canker disease on poplars trees in West Azarbayjan province by means of morphological and molecular data. For this propose, wood samples were collected from poplar and willow trees with canker symptoms during summer and autumn 2015. A total of 110 samples were collected from poplar and willow gardens in the region. Fungal spices were isolated using single spore method on water-agar (WA). A total number of 173 fungal isolates were recovered from diseases samples showing canker symptoms. Fungal isolates were identified using morphological characteristic as well as sequence data ITS-rDNA region. In order to evaluate pathogenicity of the fungal isolates, pathogenicity test was carried out in greenhouse condition and on populus alba and p. nigra. Our results revealed that several species of fungi are involved in the canker disease poplar trees in this region including Cryptosphaeria pullmanensis, Cytospora chrysosperma and Cytospora sp.. Cryptosphaeria pullmanensis was the dominant species amongst the fungal groups involved in this region with 85 percent frequency. This species was isolated only from the poplars. The results obtained by the inoculations confirmed that all the fungal isolates were pathogenic. Isolates Cytospora sp. showed maximum of the wound length on P. nigra with the size 22.75 cm and minimum on P. alba with the size 6 cm. It seems that the sensitivity of P. nigra to Cytospora species is much greater than p. alba. But the average lesion length on wood of P. alba created by Cryptosphaeria pullmanensis on was greater than P. nigra, as a result, it seems that P. alba is more sensitive than P. nigra
