.20 g were randomly divided into seven main groups (n=8): control, sham, EB or lysion groug (MS model), four treated groups which was treated by curcumin and linocurcumin (5?g/rat and 10?g/rat). One week after MS induction by ICV injection of EB, each of treated groups were treated by mentioned doses for 5 consecutive days. Characteristic of learning and spatial memory was assessed using Morris Water Maze for 5 consecutive days following the treatment period. After termination of behavioral testing days, animals were decapitated and the brain dissected to measure levels of oxidative stress parameters using method FRP, MDA and antioxidant enzymes such as SOD and CAT. Statistical analysis was performed using analysis of variance (ANOVA).Results: Results treatment with curcumin and linocurcumin in two doses of 5?g/rat and 10?g/rat significantly improved spatial memory in experimental models of MS (P< 0.05). Also, the level of oxidative stress parameters in the MS groups treated with curcumin and linocurcumin was significantly lower than those of control MS groups (P< 0.05). Results of this study showed that linocurcumin especially in high dose (10?g/rat) showed more improvement effect in special memory and oxidative stress level in comparison with other groups
TOPICAL NAME USED AS SUBJECT
Background and aim: Multiple sclerosis (MS) is an acute progressive disease in central nervous system (CNS) in which demyelination of neurons occurs. The aetiology of multiple sclerosis is unclear, but its pathology includes penetration of immune system agents into central nervous system, neuroinflammation, demyelination and axonal degeneration. Demyelinated plaques initiate and progress this disease. Oxidative stress plays an important role in multiple sclerosis pathogenesis. Curcumin (a hydrophobic poly phenol substance) is yellow pigment of turmeric. Curcumin is an antioxidant which has some effects included anti-inflammatory, anti-appoptotic, antinociception, antibacterial and immune system modulator. Because of curcumin antioxidant and anti-inflammatory effects, it seems that curcumin can be a potent treatment for neurodegenerative diseases like MS. It has shown that curcumin can increase neurogenesis in adult hippocampus. Curcumin has a poor bioavailability, so it uses in some complex and nano forms to increase its bioavailability. Polyunsatured fatty acids (PUFA) especially omega-6 group have antioxidant and anti-inflammatory effects which make them a potent substances for treatment MS. Accordingly, in this thesis we use curcumin and linoleic acid (an omega-6 fatty acid) complex substance (Linocurcumin), to investigate how the bioavailability, stability and effectiveness of curcumin increase, also we examine effect of linocurcumin in comparison with curcumin in special memory and oxidative stress condition.SM htiw star elam fo supmacoppih eht ni srekram sserts evitadixo fo noitaludom dna sserts evitadixo fo noitcuder hguorht ,yromem laitaps dna gninrael setaroilema yltnacifingis ,nimucruconil -mrof citetehtnys ni nimucruc fo noitcejniorcim taht dedulcnoc eb nac ti ,yduts ruo ot gnidroccA .eussit niarb ni ytilibaliavaoib sti esaercni nac ,mrof xelpmoc ni ti esilitu ,stceffe evitcetorporuen dna tnadixoitna sah nimucruc revewoH :noissucsiD
