مقایسه ی تمایل اتصال فاکتور نسخه برداری RUNX1 به دو الیگو نوکلئوتید نمایانگر پلی مرفیسم در توالی پالیندرومیک 5’HS4 ناحیه ی تنظیم لوکوس بتا

مطالعات قبلی ما (مرکز تحقیقات ژنتیک دانشگاه علوم بهزیستی و توانبخشی) یک رابطه ی پیوستگی بین شدت فنوتیپ بالینی بیماری بتا تالاسمی و ناحیه ی پالیندرومی پلی مرف 5’HS4 در ناحیه ی تنظیم لوکوس خوشه ی بتا گلوبین نشان داد. علاوه بر این یافته، یک موازنه ی پیوستگی نیز بین این ناحیه و ناحیه ی Xmn1-HBG2 در جمعیت بیماران مشاهده گردید. بر این اساس پیشنهاد شد نقش بارز ایجاد تغییر در فنوتیپ که به Xmn-HBG2 نسبت داده شده توسط عوامل پیوسته به Xmn1 در LCR ایفا می گردد. حال این سوال مطرح است که پلی مرفیسم در ناحیه ی چگونه می تواند در فنوتیپ بیماران تاثیر گذار باشد. در این تحقیق برای بررسی اهمیت عملکردی پلی مرفیسم در این ناحیه، تاثیر این پلی مرفیسم در اتصال فاکتورهای نسخه برداری مورد بررسی قرار گرفت. بررسی نرم افزاری در مطالعه یپیشین ما یک ناحیه ی اتصال برای RUNX1 در این ناحیه هنگامی که آلل پلی مرف در مرکز پالیندروم (TGGGG(A/G)CCCCA) آلل “A” و نه آلل “G” باشد پیش بینی کرد. این الگوی اختصاصی به آلل با نتایج آزمایشات طرح حاضربا استفاده از روش Electromobility Shifitng Assay تایید شد. پروتئین RUNX1 یک تنظیم کننده ی شناخته شده در پروسه ی خونسازی می باشد. بنا بر این نتایج حاضر پیشنهاد می کند که رل تغییر فنوتیپ 5’HS4 که در مطالعات قبلی مشاهده شده است، احتمالا به تعامل آلل -اختصاصی بین 1 RUNX و این ناحیه ارتباط دارد. این یافته ها مطالعات بیشتر برای تایید این تعاملات در vivo و بررسی پیامد های عملکردی و فنوتیپی اتصال مختص به آلل RUNX1 به ناحیه ی پالیندرومی پلی مرف 5’HS4 را پیشنهاد می نماید.
In our previous studies on the Iranian beta thalassemia patients, we identified an association between the severity of beta thalassemia phenotype and the polymorphic palindromic site at 5’ hypersensitive site 4-locus control region (5’HS4-LCR) of beta globin gene cluster. Furthermore, a linkage disequilibrium was observed between this region and XmnI-HBG2 in the patient population. Based on this data, it was suggested that the well-recognized phenotype-ameliorating role assigned to positive XmnI, could be associated with its linked elements in LCR. In here, to investigate the functional significance of polymorphism at 5’HS4-LCR, we studied its influence on binding of transcription factors. Web based predictions of transcription factor binding, revealed a binding site for RUNX1 transcription factor, when the allele at the center of the palindrome (TGGGG(A/G)CCCCA) was “A” but not when it was “G”. We confirmed this putative allele specific pattern of binding with an electromobility shifting based competition assay. RUNX1 is a well know regulator in hematopoiesis. Therefore, these data suggest that the phenotype modifying role of 5’HS4-LCR polymorphism, observed in our previous studies, could possibly be related to the allele specific binding of RUNX1 to that region. These preliminary data suggest the importance of further studies to investigate the functional and phenotypical consequences of RUNX1 allele specific binding to the polymorphic palindromic site at 5’HS4.
