.Insulin- like growth factors (IGF-I, II) are necessary for growth, development and metabolic activities of mammals, and mediate most effects of the growth hormones. Their biological activities are inhibited by binding proteins, IGFBP-s which in turn inhibits growth of cells. Different types of IGFBP-s differ in their molecular weight, location effect of their amino- acids, distribution in biological liquid and their effect on IGF activity. The objective of this study was to detect of polymorphism in IGFBP-۳ gene of the Holstein cows and it's relation to productive traits. To achieve this, blood samples were collected from ۱۰۰ Holstein cows in Dashet- e- Azarnegin dairy farm which their pedigree as well as required records were available. After DNA extraction, the PCR- RFLP technique followed by amplifying a fragment of the gene (۳۱۶ bp in length) using specific primers. Then the PCR product was digestive using restriction enzymes HaeIII and BfaI and finally electrophoresis was done on ۲ agaros gel. The digestion of PCR products with each enzyme, produced four fragments sized ۱۹۵, ۶۹, ۴۴, and ۸ bp respectively which were similar in all of studied animals.
