.Tomato Bacterial canker disease, firstly reported in Iran from Urmieh in West Azarbaijan province in ۱۹۸۸. The disease causes lesion (canker), wilting and dryness of plants, leaf and fruit spots and whole plant decline. In this study, about ۱۶۰ samples of infected plants were collected and sixty-four bacterial isolates were obtained from the infected tomato plants in West Azarbaijan during ۲۰۰۴-۲۰۰۵. All isolates identified as Clavibacter michiganensis subsp. michiganensis based on morphological, biochemical characters and pathogenecity test. An isolate of Cmm (NCPPB۳۸۲) received from Prof. Dr. Rudolf Eichenlaub Universitaet Bielefeld was used as standard. Isolates showed considerable differences in using different sources of carbon (Diglucose, Dimannose, Dimlzitose, Monitol, Diarabinose, Difructose, Sucrose, Rafinose, Alfamethyl Diglucose and Acetate). Fifteen isolates (۱۱ from West Azarbaijan, three from Golestan and the standard) selected based on phenotypic tests, and then their electrophoretic (SDS-PAGE) profiles of soluble cell proteins were studied. Most differences among the isolates, observed in ۰.۸, ۱.۵, ۲.۸, ۴.۵ and ۵.۵Rf. Plasmid extraction carried out from the selected and standard isolates. Agarose gel Electrophoresis indicated that all the isolates have two plasmids pCM۱ (۲۷.۵ kb) and pCM۲ (۷۲ kb) and no differences were found among the isolates. DNA fingerprinting using rep-PCR (BOX primer) studied among ۱۱ isolates of Cmm (eight isolates from West Azarbaijan , two from Golestan and the standard ) that were selected based on biochemical characters and protein profiles. A cluster analysis dendrograms obtained based on information derived from biochemical characters, protein electrophoresis and rep-PCR reactions, using software NTSYS-pc version ۲/o۲e, UPGMA method and Jaccard coefficient. According to the site of cut-off line, three groups(clusters) with ۷۷ similarity based on biochemical data , ۵ groups with ۹۳ similarity based on SDS-PAGE results , and six groups with ۶۹ similarity based on rep-PCR reactions were realized. Comparing the rep-PCR dendrogram with two other ones (biochemical tests and SDS-PAGE) indicated a high correlation and complementarirty. Results revealed I) difference between Iranian isolates and the standard Cmm (NCPPB۳۸۲), II) similarity between Golestan and West Azarbaijan isolates, and III) presence of different populations (strains) among the isolates selected from different areas. This is the first investigation since ۱۹۹۸ in studying genetic diversity among geographical populations of Clavibacter michiganensis subsp. michiganensis and the first report of occurring tomato bacterial disease in Golestan province-Iran
