High performance liquid chromatographic separation of some psychotherapeutic benzodiazepines with chemometrical optimization

In this study, an accurate, selective, sensitive and nondestructive high performance liquid chromatographic (HPLC) method for the separation and quantitative determination of a mixture of some 1,4 benzodiazepines (namely Nitrazepam, Diazepam and Medazepam) was developed. Using the modified simplex algorithm, together with a suitable chromatographic response function (CRF), an optimum condition of 89% CH3CN, 11% 0.1 M acetate buffer, 3.44 ml/min flow rate, and 5 mu1 injected volume were obtained. Under these conditions the separation was achieved in less than two minutes. The effect of the experimental variables on the CRF was studied. The method was found to be suitable for the determination of these compounds in proprietary drugs without suffering interferences from excepients. A recovery of 101.6%, an accuracy of 0.11 ppm and a detection limit of 200 ppb were obtained. The developed method was superior to the existing standard method described for the determination of diazepam in drug tablets.
